Abstract:Objective: To observe the apoptosis of myeloidderived suppressor cells (MDSCs) that is treated by IL17 and explore the effect of IL17 on apoptosis of MDSCs. Methods: Lewis lung carcinoma (LLC) tumor bearing models were constructed with subcutaneous injection of Lewis cells. MDSCs were isolated from spleen in tumorbearing mice and cultured in RPMI 1640 culture media. After treating MDSCs with IL17 for 24 h, apoptosis of MDSCs were detected by Annexin V and PI. Furthermore,Western blotting was also used to detect the expression of antiapoptotic protein Bcl2 after treating MDSCs with IL17 for 6 h. Results: The counts of viable cells of MDSCs were(4.123±0.259 8)×104 in control group and (9.547±0.962 6)×104 in IL17 treatment group (P<0.01); percentage of viable cells of MDSCs was (45.33±1.556)% in control group and (59.55±2.831)% in IL17 treatment group (P<0.05). The later apoptosis of MDSCs were (22.87±1.349)% in control group and (13.70±1.003)% in IL17 treatment group(P<0.01). Results of Western blotting showed that the expression level of antiapoptotic protein Bcl2 significantly increased after IL17 treatment(P<0.05). Conclusion: IL17 may inhibit the apoptosis of MDSCs in vitro significantly.
