Abstract:Objective: To investigate the expression and clinical significance of unconventional prefoldin RPB5 interactor(URI) in gastric cancer tissues as well as the effect of URI on the apoptosis of gastric cancer cells treated with cisplatin. Methods: Immunohistochemistry was used to detect the expression of URI in tissue microarray of gastric cancer. MGC803 and SGC7901 gastric cancer cells were cultivated in vitro. Hiperfect transfection reagents were used to transfect URI siRNA interference fragment into the two strains of gastric cancer cells respectively. The expression of URI in gastric cancer cells was detected by qRTPCR and Western blotting. Gastric cancer cells was divided into blank group, URI siRNA group and cisplatin induced untransfected group, scrambled group, URI siRNA group. Flow cytometry was used to detect the apoptosis of gastric cancer cells treated with cisplatin. Results: The expression of URI in cytoplasm of gastric cancer tissue was significantly higher than that in the adjacent tissues (P<0.05). The URI expression of cell nucleus in gastric cancer tissue was higher than that in paracancerous tissue, but the difference was not statistically significant. The expression of URI was not significantly correlated with the survival period and clinical stage of gastric cancer patients. The apoptosis rates of gastric cancer cells with URI knockdown were increased significantly compared to scrambled groups(P<0.01). Conclusion: As an oncoprotein, URI helped inhibitting apoptosis of gastric cancer cells and may play a role in gastric carcinogenesis.
