369 Stemness-enhanced mouse embryonic fibroblast-derived extruded vesicles inhibit multiple tumor cell growth

Objective: To explore the effect of small molecule compound combined with 3D culture on the stemness of mouse embryonic fibroblast (MEF), and the effect of extruded vesicles on the growth of different tumor cells. Methods: First, MEFs were treated with different concentrations of small molecule drugs β-nicotinamide mononucleotide (NMN), betaine, rapamycin and vitamin C under 2D culture conditions, and Oct-4  mRNA expression level was detected by realtime quantitative PCR, and the optimal concentration for promoting cell stemness was screened out; 2, 3, 4 combinations of 4 drugs were used to treat MEF according to the optimal concentration, and the optimal combination was screened. Secondly, the selected chemical combinations were combined with 3D culture, and the mRNA and protein expression levels of stemness indicators Oct-4 and Nanog were detected by realtime quantitative PCR and Western blotting, respectively, and the optimal combination for promoting cell stemness was screened out. Finally, MEFs were prepared into vesicles by extrusion method, and Hepa1-6, Lewis and GL261 tumor cells were treated with vesicles, and the effect of vesicles on the growth of several tumor cells and MEFs was detected by CCK8 method, respectively. Results: The optimal concentration of NMN, betaine and rapamycin on MEF stemness promotion was 10 μmol/L, and the optimal concentration of vitamin C was 100μmol/L; the combination of NMN+betaine had the strongest effect on promoting MEFs stemness. Compared with the 2D culture group, the expression level of Oct-4 mRNA in the NMN+betaine combined with 3D culture group was significantly increased by nearly 600 times (P<0.05). The results of Western blotting showed that the expression levels of Oct-4 and Nanog in the combined 3D culture group of betaine+vitamin C, NMN+betaine, rapamycin+betaine, ect. were significantly increased, and the expression level in NMN+betaine combined 3D culture group was the highest (P<0.05). The extruded vesicles of NMN+betaine combined with 3D culture group greatly inhibited the growth of mouse hepatoma cells Hepa1-6, glioma cells GL261 and lung cancer cells Lewis (P<0.05), but the inhibitory effect on MEFs was not obvious. Conclusion:NMN+betaine combined with 3D culture could significantly enhanced the stemness of MEFs, and its extruded vesicles could markedly inhibit the growth of Hepa1-6, Lewis and GL261 tumor cells.

376 Inhibition of human umbilical cord mesenchymal stem cells derived exosomes on the capillarization of liver sinusoidal endothelial cells#br#

[Abstract]Objective: To investigate the effect of human umbilical cord mesenchymal stem cell derived exosomes(MSC-Ex) on the capillarization of liver sinusoidal endothelial cells(LSECs). Methods: Mesenchymal stem cells were isolated from human umbilical cord tissue and cultured. Subsequently, exosomes were extracted from culture supernatant by ultracentrifugation. The morphology and characterization of exosomes were identified by transmission electron microscopy, nanoparticle tracking analysis and Western blotting. LSECs were vascularized by TNF-α in vitro, and the uptake of MSC-Ex by LSEC was observed by co-culture with red fluorescent dye DIO labeled MSC-Ex. QRT-PCR and Western blotting were used to detect the mRNA and protein expression of angiopoietin-2 (Ang-2) and CD34 in LSECs, respectively, and the tube forming ability of LSEC was detected by tubule formation test. The hepatic fibrosis model of ICR mice induced by methionine and choline deficiency diet (MCD) was established, MSC-Ex (10 mg/kg) and 100 μL PBS was injected via tail vein, once every 3 days, for a total of 5 injections. Collagen deposition and Ang-2 protein expression in liver tissue were observed by Sirius red staining and detected by immunohistochemistry, respectively. Results: Western blotting results showed that MSC-Ex expressed characteristic markers Alix, TSG101, CD63 and CD9. Transmission electron microscopy indicated that MSC-Ex was a diskshape vesicle with diameters of about 100 nm. Nano-particle analyzer showed that the particles number of exosome was about 3.4×1011/mL. MSCEx could be ingested by LSECs, and could significantly inhibited Ang2 and CD34 expression and tube formation of LSECs induced by TNF-α. Sirius red staining and immunohistochemistry indicated that MSC-Ex could inhibit collagen deposition and the protein expression of Ang-2 in fibrotic liver tissue. Conclusion:Human umbilical cord MSC-Ex could inhibit the capillarization of LSEC in vivo and in vitro.

385  Uric acid promotes the proliferation and migration of prostate cancer cell through down-regulating the expression of lncRNA MIR22HG#br#

[Abstract]Objective: To investigate whether uric acid (UA) regulates the proliferation and migration of prostate cancer cells through lncRNA MIR22HG. Methods: Prostate cancer DU145 cells were divided into control group and UA (400 μmol/L) treatment group, transfected with siMIR22HG or siNC group, and transfected with pcDNAMIR22HG or pcDNA group, and UA+pcDNA-MIR22HG or UA+pcDNA group. The expression of MIR22HG in DU145 cells was detected by qRT-PCR, the cell proliferation ability was detected by CCK-8, the expression of cell cycle related factors (Cyclin D1, CDK4 and P21) was detected by Western blotting, and the cell migration ability was measured by Transwell. Results: Compared with the control group, after being treated with 400 μmol/L uric acid, the expression of MIR22HG in DU145 cells decreased significantly (P<0.01), the viability and migration ability of prostate cancer DU145 cells increased significantly (P<0.01), the expression levels of Cyclin D1 and CDK4 protein increased significantly (P<0.01), and the expression level of P21 protein decreased significantly (P<0.01). Compared with the corresponding control group, downregulation of MIR22HG could significantly promote the proliferation and migration of prostate cancer DU145 cells (P<0.01), while upregulation of MIR22HG could inhibit their proliferation and migration (P<0.01). At the same time, overexpression of MIR22HG could reverse the promoting effect of uric acid on the proliferation and migration of DU145 cells (P<0.01). Conclusion: Uric acid can promote the proliferation and migration of prostate cancer cells, which may be attributed to the down regulation of MIR22HG expression.

391 Bioinformatics exploration on the mechanism of osteoarthritis miRNA-mRNA regulatory network

[Abstract]Objective: Based on bioinformatics, the aim of this study is to explore the mechanism of osteoarthritis miRNAmRNA regulatory network, the Hub genes would be screened and its molecular regulatory mechanism in osteoarthritis is also analysed. Methods: Through the GEO database, the miRNA microarray dataset (GSE105027) and the mRNA microarray dataset (GSE55235) of osteoarthritis were obtained, and the differentially expressed genes were analyzed. Then FunRich software was used to predict the transcription factors of differentially expressed miRNAs, functional enrichment analysis and target gene prediction, and then the predicted target genes were crossed with the differentially expressed mRNAs to obtain the miRNAmRNA regulatory network. Then the STRING database and Cytoscape software were used to construct a protein interaction network and screen out the Hub genes. Finally, R language was used to analyze the function and related pathways of Hub gene. Results: ① A total of 265 miRNAs and 838 mRNAs were shown differentially expressed; ② The prediction of 265 miRNAs resulted in a total of 203 transcription factors, of which EGR1 had the most significant difference. Enrichment analysis showed that miRNAs were mainly involved in the regulation of nucleobase, nucleoside, nucleotide and nucleic acid metabolism, cell growth regulation, transcription and other processes; ③ A total of 3 572 target genes were predicted, and the screening was based on the negative regulatory relationship between miRNA and mRNA. And 96 mRNAs and 25 miRNAs with negative regulation of cross-expression were identified; ④ JUN, FOS, IL-6, TNF, IL-1β, and CXCL8 are the Hub genes in the protein interaction network; ⑤ The enrichment analysis mainly covers biological processes such as the response to lipopolysaccharide, the response to bacteria-derived molecules, and the regulation of DNAbinding transcription factor activity, involving IL-17, Toll-like receptors, AGERAGE, TNF and NODlike receptors signaling pathways. Conclusion: The miRNAmRNA regulatory network of osteoarthritis was successfully screened and constructed, and the molecular regulation mechanism of miRNA and its target genes in the occurrence and development of osteoarthritis was clarified to a certain extent.

398 Preparation of experimental animal model of mouse deep vein thrombosis#br#

Objective: To prepare the experimental animal model of deep vein thrombosis(DVT) in mice. Methods: Fiftysix C57BL/6J mice were randomly divided into five groups: normal control group, sham operation group, operation group 1, operation group 2 and endotoxin+operation group. In operation group 1, 90% of inferior vena cava were ligated. The sham operation group performed the same operation except ligation. Endotoxin+operation group and operation group 2 were intraperitoneally injected with endotoxin and equal volume of normal saline 2 hours before surgery, and the other operations were the same as operation group 1. The general situation of the mice before and after the inferior vena cava stenosis was evaluated. The images of the structures and vessels around the inferior vena cava were obtained by Doppler ultrasonography before and immediately after the operation. The length, weight and percentage of thrombosis were determined 48 h after the operation as the quantitative indexes to evaluate the DVT model. Results: The total mortality after inferior vena cava stenosis was 25.0% and the success rate was 70.8%. Immediate postoperative ultrasound showed that there was still venous blood flow through the ligation site. No thrombus was observed in the normal control group and the sham operation group, and there was no significant difference in the size of thrombus between operation group 1 and operation group 2 (P>0.05). Compared with the operation group 2, the length of thrombus in the endotoxin+operation group was significantly increased (P<0.05), and the weight of thrombus was increased, but the difference was not significant (P>0.05). Conclusion: The model of DVT in mice prepared in this study has good operability, validity and practicability.

403 Analysis of correlation between peripheral blood inflammatory markers and the number of significant intracranial atherosclerotic stenosis in elderly patients with acute ischemic stroke

Abstract]Objective: To detect the changes of inflammatory markers in peripheral blood of elderly patients with acute ischemic stroke(AIS) and analyze their correlation with the number of significant atherosclerotic stenosis(SAS) in the brain.Methods: Patients elder than 60 years with AIS of large artery atherosclerosis and small artery occlusion subtypes hospitalized in Department of Neurology, Affiliated Hospital of Jiangsu University from August 1st, 2021 to December 31th, 2021 were consecutively enrolled in the study. Biochemical indicators were tested by laboratory examination. Peripheral inflammatory indices were collected via routine blood tests, including leukocyte count, neutrophil count, monocyte count, lymphocyte count, and neutrophiltolymphocyte ratio(NLR). Intracranial SAS was evaluated by magnetic resonance angiography(MRA), which was defined as ≥50% stenosis or occlusion. The subjects were divided into three groups based on the number of intracranial SAS: no SAS(n=77), single SAS(n=45) and multiple(≥2) SAS group(n=33); and the clinical characteristics including biochemical indicators and inflammatory markers were compared among the three groups. Multivariate logistic regression was used to analyze the correlation between the number of intracranial SAS and inflammatory markers. According to the most severe stenosis of intracranial SAS, 78 subjects with SAS were divided into 3 groups: moderate stenosis group(n=23), severe stenosis group(n=17) and occlusion group(n=38). Inflammatory markers of three groups were compared. Spearman rank correlation was used to analyse the correlation between inflammatory indexes and the number of intracranial SAS. Results: Compared with the nonSAS and/or singlevessel SAS group, the age, white blood cell count, neutrophil count and NLR in the multivessel SAS group were significantly increased(all P<0.05). Multivariate logistic regression analysis showed that the number of intracranial SAS was independently correlated with high white blood cell count, high and medium neutrophil count, and high NLR(all P<0.05). There was no significant difference in the inflammatory indexes between the moderate stenosis, severe stenosis and occlusion groups(all P>0.05). The results of Spearman correlation analysis showed that white blood cell count and neutrophil count were weakly correlated with the number of intracranial SAS(r=0.255, P=0.024; r=0.319, P=0.004).Conclusion: Peripheral blood inflammatory indicators in elderly patients with AIS showed increased white blood cell count, neutrophil count and NLR, which were weakly correlated with the number of intracranial SAS.

409 The value of ultrasonic images texture analysis in differential diagnosis of parotid pleomorphic adenoma and adenolymphoma#br#

Objective: To investigate the value of ultrasonic images texture analysis in differential diagnosis of parotid pleomorphic adenoma and adenolymphoma. Methods: A total of 43 parotid pleomorphic adenomas and 54 adenolymphomas were selected. The ultrasonic images were imported into Mazda 4.6 software and ROIs were manually drawed. The optimum texture parameters were selected through mutual information (MI), Fisher coefficient, probability of classification error and average correction coefficient (POE+ACC) and the combination of three methods (Fisher+POE+ACC+MI), respectively.The model of artificial neural network was built. The assessment results of texture analysis and ultrasonic doctors were compared with the pathology. And misdiagnostic rates were analyzed between texture analysis and ultrasonic doctors. Results: The age and proportion of male in patients with parotid adenolymphoma were higher than those with parotid pleomorphic adenoma,there were statistically difference (P<0.001). In ultrasonic features, there were statistical difference between cystic degeneration and blood flow in two groups (P<0.001 or P<0.05).Among the 30 groups of texture parameters extracted by Mazda, there were 8 groups with statistical differences (all P<0.05).The misdiagnostic rates of MI, POE+ ACC, Fisher and Fisher+POE+ ACC+MI were 53.61%(52/97), 44.32%(43/97), 21.65%(21/97), 17.53%(17/97), respectively. Furthermore, the misjudgment rate of the combination of three methods was lower than that of the ultrasound doctors(29.90%, 29/97), there was statistical differences(χ2=4.103, P<0.05). Conclusion:Ultrasonic images texture analysis can be used to differentiate parotid pleomorphic adenoma and adenolymphoma.

415 The predictive value of changes in QRS duration after cardiac resynchronization therapy in all-cause mortality and sudden cardiac death

Objective: To evaluate the predictive value of changes in QRS duration (ΔQRSd) after cardiac resynchronization therapy (CRT) in allcause mortality and sudden cardiac death. Methods: Patients underwent CRT between March 2011 and December 2020 in Wuxi People′s Hospital Affiliated to Nanjing Medical University were enrolled. The clinical data including 12lead electrocardiography were obtained. QRS durations derived from pre and postimplantation electrocardiography were measured. Predictors for allcause mortality and sudden cardiac death were evaluated using Cox regression analysis. Xtile software was used to analyze the cutoff of ΔQRSd for predicting allcause mortality and sudden cardiac death. ΔQRSd was assessed in relation to allcause mortality and sudden cardiac death by KaplanMeier survival analysis. Results: Sixtysix patients were enrolled with a median followup time of 72 months, 15 died and 16 suffered from sudden cardiac death. ΔQRSd was a predictor for allcause mortality in multiple Cox regression analysis(HR=1.019, 95% CI:1.004-1.035,P=0.014). Both ΔQRSd(HR=1.016, 95% CI:1.002-1.031, P=0.024) and complete left bundle branch block(HR=0.211, 95% CI:0.065-0.682, P=0.009) were predictors for sudden cardiac death. The cutoff of ΔQRSd for predicting allcause mortality and sudden cardiac death was 0 ms. The preimplantation left ventricular enddiastolic diameter in the ΔQRSd> 0 ms group was significantly higher than that in the ΔQRSd ≤ 0 ms group (P<0.001), and the preimplantation QRS duration was significantly lower than that in the ΔQRSd ≤ 0 ms group (P=0.001). KaplanMeier survival analysis revealed that ΔQRSd> 0 ms was a predictor of allcause mortality, sudden cardiac death, 5year survival rate, and 5year sudden cardiac death rate after CRT (P<0.001, <0.001, 0.01 and <0.001, respectively). Conclusion: A postimplantation widening in QRS duration is an independent predictor of allcause mortality and sudden cardiac death after CRT.

422 Expression and significance of the long noncoding RNA TMEVPG1 in peripheral blood from sepsis patients#br#

Objective: To investigate the expression of long noncoding RNA TMEVPG1 in peripheral blood mononuclear cells(PBMCs) from sepsis patients and the corresponding clinical significance. Methods: A total of 31 sepsis patients admitted to the Department of Critical Care Medicine of the Affiliated People′s Hospital of Jiangsu University from January 2021 to December 2021 were selected as the sepsis group, and 32 healthy subjects as the control group. The transcript levels of TMEVPG1 and interferonγ(IFNγ) in PBMCs were detected by realtime quantitative PCR. The relationship between TMEVPG1 levels and IFN-γ mRNA levels, APACHE Ⅱ score, neutrophil to lymphocyte ratio(NLR), and neutrophil to lymphocyte and platelet ratio(N/LPR) were analyzed. ROC curve was performed to analyze the diagnostic value of TMEVPG1 for sepsis. Results: The transcript levels of TMEVPG1 in PBMCs from sepsis patients were significantly decreased compared with the controls. There was a significantly inverse correlation between TMEVPG1 levels and APACHE Ⅱ score of sepsis patients. Meanwhile, TMEVPG1 expression also negatively correlated with NLR and N/LPR in patients with sepsis. Compared with the controls, IFN-γ mRNA expression in PBMCs from sepsis patients was downregulated, and there was a significantly positive correlation between TMEVPG1 levels and IFN-γ mRNA levels in sepsis patients. ROC curves showed that TMEVPG1 could distinguish sepsis patients from healthy controls. Conclusion: Our findings indicated that TMEVPG1 was decreased in the peripheral blood of sepsis patients and might play a pivotal role in the pathogenesis of sepsis by regulating the expression of IFN-γ.

427 Application of body composition assessment in diagnosing malnutrition among systemic lupus erythematosus#br#

Objective: To evaluate the nutritional status of patients with systemic lupus erythematosus(SLE) by body composition assessment and to analyze the risk factors of malnutrition in SLE patients. Methods: A total of 120 patients with SLE were selected. According to the global leadership initiative on malnutrition(GLIM), the SLE patients were divided into nondystrophic group (n=63) and dystrophic group(n=57). The clinical data, laboratory indicators, body composition data, and disease activity (SLEDAI2000 score) among the groups were compared and risk factors of malnutrition in SLE patients were analyzed by Logistic regression analysis.Results: ① The levels of height, body weight, hemoglobin, serum albumin, complement C3, and complement C4 in the SLE dystrophic group were statistically lower than those in the non-dystrophic group(P<0.05). The median of the SLEDAI2000 score of the SLE dystrophic group was 6.5, which was significantly higher than that of the non-dystrophic group(1.0, P<0.001). ② The wholebody phase angle, total body water, protein, skeletal muscle, body cell volume,intracellular fluid of the SLE dystrophic group were significantly lower than those in the non-dystrophic group (P<0.05).③ The Logistic regression analysis showed whole body phase angle (OR=0.364, 95% CI: 0.180-0.737, P=0.005), protein(OR=0.195, 95% CI: 0.042-0.892, P=0.035) were independent influencing factors of malnutrition in SLE patients. Conclusion: Using body composition assessment to evaluate the nutritional status of SLE patients can comprehensively understand the nutritional status of patients, which has certain clinical significance for multidisciplinary management of SLE.

433 Correlation of waisthip ratio with glucose and lipid metabolism, insulin-like growth factor1 and insulin resistance in patients  with polycystic ovary syndrome

Objective: To study the correlation of waisthip ratio with glucose and lipid metabolism, insulinlike growth factor1(IGF1) and insulin resistance(IR) in polycystic ovary syndrome(PCOS) patients. Methods: A total of 110 PCOS patients who were admitted to Jiangsu Shengze Hospital Affiliated to Nanjing Medical University from January 2018 to September 2021 were selected as the PCOS group, and 60 healthy subjects during the same period as the control group. Waisthip ratio, glucose and lipid metabolism indexes, IGF1 and homeostasis model for IR (HOMAIR) were measured in the two groups. The glucose and lipid metabolism indexes included fasting blood glucose(FPG), 2 h postprandial blood glucose (2 h PBG), total cholesterol(TC), highdensity lipoprotein cholesterol(HDLC), lowdensity lipoprotein cholesterol(LDLC), triglyceride(TG) and fasting insulin (FINS). The patients with waisthip ratio≥0.80 in the PCOS group were subgrouped into the abdominal obesity group, and the patients with waisthip ratio <0.80 into the nonabdominal obesity group, and the glucose and lipid metabolism indexes, IGF1, and HOMAIR were compared between the two groups.Pearson correlation analysis was used to evaluate the correlation between waisthip ratio and glucose and lipid metabolism, IGF1 and IR in PCOS patients. The ROC curve was used to analyze the predictive value of waisthip ratio for abnormal glucose and lipid metabolism, IGF-1 and IR in PCOS patients. Results: Compared with the control group, the PCOS group showed higher levels of the waisthip ratio, 2 h PBG, LDL-C, TG, FINS, IGF-1, HOMA-IR, and lower level of HDL-C (all P<0.05), while there was no significant difference between the levels of FPG and TC (both P>0.05). Compared with the nonabdominal obesity group, the levels of 2 h PBG, LDLC, TG, FINS, IGF-1 and HOMA-IR in the abdominal obesity group were significantly increased, and the levels of HDL-C were significantly decreased (all P<0.05), while there was no statistically significant difference between the levels of FPG and TC (both P>0.05). In PCOS patients, waisthip ratio was positively correlated with 2 h PBG, TG, IGF-1, HOMA-IR (all P<0.05), while negatively correlated with HDL-C(P<0.05). Waisthip ratio had good predictive value for abnormal blood glucose metabolism, abnormal blood lipid metabolism and IR in PCOS patients(AUC were 0.821, 0.793, 0.782, respectively). Conclusion: PCOS patients, especially those with higher waisthip ratio, showed obvious glycolipid metabolism disorders, abnormal increase of IGF1 and IR, and waisthip ratio has a good predictive value for glycolipid metabolism disorders and IR in PCOS patients.

439 Effects of intraoperative goaldirected fluid therapy on postoperative rehabilitation of patients undergoing laparoscopic sleeve gastrectomy#br#

Objective: To compare the postoperative rehabilitation effect of the goaldirected fluid therapy (GDFT) and the restrictive fluid therapy (RFT) for obese patients undergoing laparoscopic sleeve gastrectomy. Methods: Sixty obese patients who underwent elective laparoscopic sleeve gastrectomy under general anesthesia from May to October 2021 in the Affiliated Hospital of Xuzhou Medical University were selected and divided into RFT group (group RFT) or GDFT (group GDFT) according to the random number table method, with 30 patients in each group. The fluctuation of heart rate and mean arterial pressure in group RFT did not exceed the preoperative baseline 20%. Group GDFT used the FloTrac/Vigileo monitoring system to monitor stroke volume variation (SVV), maintaining SVV≤13%. The incidence of postoperative complications in the two groups was observed. Between the two groups, the use of intraoperative vasoactive drugs and the incidence of intraoperative hypotension were compared; the first postoperative anal exhaust time was compared. The scores of gastrointestinal quality of life index (GIQLI) before and after surgery at 2, 5, 10, 16 weeks were compared. Results: Compared with group RFT, the incidence of postoperative complications in group GDFT was significantly reduced (P<0.05), the utilization of intraoperative vasoactive drugs and the incidence of intraoperative hypotension were also significantly lower (both P<0.05), the first postoperative anal exhaust time was significantly earlier (P<0.01). The GIQLI score in group GDFT was significantly higher than that in group RFT at 2, 5 and 10 weeks after surgery (all P<0.05). Conclusion: The goaldirected fluid therapy based on the FloTrac/Vigileo can promote postoperative rehabilitation of obesity patients.

444 Screening and activity verification of osteogenic compounds from Salvia miltiorrhiza based on virtual screening#br#

Objective: To screen bonepromoting compounds from Salvia miltiorrhiza by molecular docking technology and verify their activity in vitro. Methods: TCMSP database was used to query the active components of Salvia miltiorrhiza. Molecular docking was performed by Autodock vina. Cell proliferation was investigated by CCK-8 assay. Alkaline phosphatase (ALP) activity test, DAPI-Photopeptide double staining and alizarin red staining were used to observe the promoting effect on bone. Results: Four active components, including danshensu, salvianolic acid B, tanshinone ⅡA and cryptotanshinone, were obtained by preliminary screening. CCK8 results showed that salvianolic acid B could promote the proliferation of preosteoblasts in MC3T3-E1 mouse preosteoblast cells. ALP activity assay showed that danshensu and salvianolic acid B significantly increased ALP activity. The results showed that salvianolic acid B could promote bone formation in MC3T3-E1 cells by DAPI-Phopeptide double staining and alizarin red staining. Conclusion: Salvianolic acid B screened by molecular docking technology is proved to exert certain osteogenic effect after activity verification.

452 Simultaneous determination of eight components in Prunella vulgaris L. by dual wavelength HPLC#br#

Objective: To establish a HPLC method for simultaneous determination of caffeic acid, pcoumaric acid, scopoletin, rosmarinic acid, esculetin, rutin, hyperin and quercetin in Prunella vulgaris L.. Methods: The separation was performed on a InertSustainTM ODSC18 column (250.0 mm×4.6 mm, 5 μm), with mobile phase consisted of acetonitrile-0.1% acetic acid solution flowing at 1.0 mL/min in a gradient elution manner. The column temperature was 30 ℃ and the detection wavelength was set at 325 nm and 360 nm. The established HPLC method was investigated by linear relationship test, precision test, stability test, repeatability test and recovery test. Results: The eight components showed good linear relationship in the range of 1-100 μg/mL, and the average recoveries were from 98.63% to 102.36%. Relative standard deviations of precision test, stability test and repeatability test were all lower than 3%. Conclusion: This simple and reliable method may be applied to accurately determine the contents of eight components, which could be used for the quality control of Prunella vulgaris L..
