Abstract:Objective: To explore the effect of cyclophilin A on the autophagy activation in RAW264.7 cells, one of the murine microphage line. Methods: RAW264.7 cells were incubated with various concentrations of cyclophilin A(0,1,10,100,500,1000 ng/mL) to figure out the optimal nontoxic concentrations of cyclophilin A for subsequent test. Western blotting was used to detect the expression of autophagy-related proteins LC3 Ⅰ and LC3Ⅱ. The RAW264.7 cells were treated with 100 ng/mL cyclophilin A for 6 hours, and then number of autophagosome was observed by fluorescence microscopy and transmission electron microscopy. Results: With the increasing of cyclophilin A concentration autophagy was gradually induced and reached the peak at the concentration of 100 ng/mL. Compared with control group, the number of autophagosomes significantly increased according to fluorescence and transmission electron microscopic observation. Conclusion: Cyclophilin A could induce autophagy in RAW264.7 macrophages.
