Abstract:Objective: To approach the influence and mechanism of radiotherapy combined with resveratrol on apoptosis of human esophageal squamous carcinoma TE-10 and KYSE-150 cells. Methods: The influence of various concentrations of resveratrol (0, 25, 50,75 and 100 μmol/L) on proliferation of TE10 and KYSE150 cells were detected by CCK8, which was used to figure out the optimal resveratrol concentration, then the proliferation of TE-10 and KYSE-150 cells treated with resveratrol combined with different radiation doses were tested by CCK8. The clone formation was used to detect the radiosensitivity of cells with different treatment. Cell apoptosis was observed by FACS in the following groups: control, intervened with resveratrol only, intervened with radiation dose 4 Gy only, intervened with radiation dose 4 Gy and resveratrol.The protein expression levels of p-H2A.X(Tyr142),p-H2A.X(Ser139), JNK1, cleaved-caspase3 were evaluated by Western blotting. Results: Resveratrol has no obviously effect on cell growth of TE-10 and KYSE-150 cells.Compared with the irradiated group and the resveratrol treatment group, the rate of number of cell colonies in the group of resveratrol combined with X-rays irradiation were remarkly increased(P<0.01). The results of flow cytometry showed that resveratrol combined with irradiation significantly promoted the apoptosis of esophageal squamous cell carcinoma compared with the irradiated group(P<0.01).Western blotting showed that the expression level of p-H2A.X(Tyr142)was significantly higher under resveratrol treatment than that of in groups without resveratrol treated, and the expression of cleaved-caspase 3 protein was up-regulated by JNK1 expression, which was further activated by radiotherapy. Conclusion: Radiotherapy combined with resveratrol can increase the apoptosis rate of ESCC after radiotherapy. Resveratrol may be a sensitizer to radiotherapy.
