Abstract:Objective: To clarify the interactions between phosphorylated Y705 and phosphorylated S727 in signal transducer and activator of transcription 3(STAT3). Methods: Mutant plasmids of STAT3 Y705F and S727A were constructed and transfected into TSC2-/- MEF cell line, respectively. The phosphorylation level of S727 in STAT3 Y705F, and the phosphorylation level of Y705 in STAT3 S727A were detected by Western blotting, respectively. Results: Gene sequencing showed that TCC were mutated into GCC and TAC were mutated into TTC. Western blotting demonstrated that Y705 and S727 have been mutated successfully and the Y705F mutation downregulated phosphorylation level of S727 in STAT3, while S727A mutation downregulated phosphorylation level of Y705 in STAT3. Conclusion: There is a synergistic effect between phosphorylated Y705 and phosphorylated S727 in STAT3.
