Abstract:[Abstract] Objective: To study the percentage of CD4+CD25+Foxp3+, CD4+IL-17+ cells and the expression Foxp3, IL-17, RORγt mRNA in peripheral blood from patients with asthma. Methods: The specimens of peripheral blood mononuclear cell(PBMC) were collected from 23 patients of asthma in remission stage, 25 patients of acute paroxysmal asthma and 20 healthy persons(control group). Intracellular flow cytometry detection of the percentage of CD4+CD25+Foxp3+ cells and CD4+IL-17+ cells was established after isolation of untouched human CD4+T cells from PBMC. The expression level of Foxp3, IL-17, RORγt mRNA in CD4+T cells was determined by realtime fluorescence quantitative RT-PCR. Results: Immunomagnetic bead shade selected CD4+T cells reached a purity of more than 90%. The percentage of CD4+CD25+Foxp3+ cells and the expression level of Foxp3 mRNA of the acute paroxysmal group was significantly lower than that of the control group and remission group(P<0.01), and the comparison of the control group and remission group didn't have significant deviation(P>0.05). The percentage of CD4+IL-17+ cells and the expression level of IL-17、RORγt mRNA of the acute paroxysmal group was significantly higher than those of the control group and remission group (P<0.01),and the expression level of IL-17、RORγt mRNA in the remission group was also significantly higher than the control group(P<0.05). Conclusion:The percentage of CD4+CD25+Foxp3+cells and the expression of Foxp3 mRNA in peripheral blood from patients with asthma decreased significantly.The percentage of CD4+IL-17+cells and the expression of IL-17、RORγt mRNA in peripheral blood from patients with athma increased significantly.The imbalance of Treg/Th17 perhaps correlated with the acute episode of asthma.
