The effect of rL-IL29 on biological characteristics of lung adenocarcinoma A549 cells and related mechanism

Abstract:Objective: To investigate the effect of recombinant Newcastle disease virus expressing IL29(rLIL29) on the proliferation, migration and invasion of lung adenocarcinoma A549 cells and explore the potential mechanism. Methods: A549 cells were cultured in vitro, then infected by rLIL29, Newcastle disease virus LoSota(NDVL) or treated by PBS. The expression of NDV HN and IL29 protein in cells was detected by Western blotting method.The effects of rL-IL29 on the proliferation, migration and invasion of lung adenocarcinoma A549 cell lines were detected by CCK8, scratch test and Transwell invasion test.The expression level of E-cadherin, MMP2, vimentin and AKT/NF-κB pathway associated proteins were detected by Western blotting. Results: NDV and IL-29 protein were stablely expressed in A549 cells infected by rL-IL29, and significantly higher than those of NDVL and control group. After infected with rL-IL29 and NDVL, the ability of proliferation,migration and invasion of A549 cells was inhibited, and rL-IL29 group is more powerful than the control group and NDVL group.The expression level of E-cadherin in the A549 cells was significantly increased after rL-IL29 infection, and the expression of vimentin,MMP2 (66 kDa/72 kDa), p-AKT and P65 protein was all significantly decreased(all P<0.05). Conclusion: rL-IL29 inhibited the proliferation,invasion and migration of lung adenocarcinoma A549 cells,and the mechanism may be up-regulation of of E-cadherin protein expression and downregulation of MMP2 and vimentin protein expression by inhibiting the AKT/NF-κB signaling pathway.
