尿酸通过下调lncRNA MIR22HG的表达促进前列腺癌细胞增殖及迁移

摘要目的: 探讨尿酸（uric acid, UA）是否通过lncRNA MIR22HG调控前列腺癌细胞的增殖和迁移。方法: 选用前列腺癌DU145细胞，将其分为对照组和尿酸处理组（400 μmol/L），转染siMIR22HG或si-NC组，转染pcDNA-MIR22HG或pcDNA组，以及UA+pcDNA-MIR22HG或UA+pcDNA组。应用qRT-PCR法检测DU145细胞中MIR22HG的表达，CCK-8法检测细胞增殖能力，蛋白质印迹法检测细胞周期蛋白D1（Cyclin D1）、细胞周期蛋白依赖性激酶4（CDK4）和P21的表达，Transwell测定细胞迁移能力。结果: 与对照组比较，400 μmol/L尿酸处理后，DU145细胞的MIR22HG表达显著下降（P＜0.01），同时前列腺癌DU145细胞活力及迁移能力均显著增强（P均＜0.01），Cyclin D1、CDK4蛋白表达水平亦显著升高（P均＜0.01），P21蛋白表达水平显著下降（P＜0.01）。与相应对照组相比，下调MIR22HG能够显著促进前列腺癌DU145细胞增殖、迁移（P均＜0.01），而上调MIR22HG可以抑制其增殖、迁移（P＜0.01）。同时，过表达MIR22HG能够逆转尿酸对DU145细胞增殖和迁移能力的促进作用（P均＜0.01）。结论:尿酸通过下调MIR22HG的表达促进前列腺癌DU145细胞的增殖和迁移。

Abstract：［Abstract］Objective： To investigate whether uric acid （UA) regulates the proliferation and migration of prostate cancer cells through lncRNA MIR22HG. Methods： Prostate cancer DU145 cells were divided into control group and UA (400 μmol/L) treatment group, transfected with siMIR22HG or siNC group, and transfected with pcDNAMIR22HG or pcDNA group, and UA+pcDNA-MIR22HG or UA+pcDNA group. The expression of MIR22HG in DU145 cells was detected by qRT-PCR, the cell proliferation ability was detected by CCK-8, the expression of cell cycle related factors （Cyclin D1, CDK4 and P21) was detected by Western blotting, and the cell migration ability was measured by Transwell. Results： Compared with the control group, after being treated with 400 μmol/L uric acid, the expression of MIR22HG in DU145 cells decreased significantly （P<0.01), the viability and migration ability of prostate cancer DU145 cells increased significantly （P<0.01), the expression levels of Cyclin D1 and CDK4 protein increased significantly （P<0.01), and the expression level of P21 protein decreased significantly （P<0.01). Compared with the corresponding control group, downregulation of MIR22HG could significantly promote the proliferation and migration of prostate cancer DU145 cells （P<0.01), while upregulation of MIR22HG could inhibit their proliferation and migration （P<0.01). At the same time, overexpression of MIR22HG could reverse the promoting effect of uric acid on the proliferation and migration of DU145 cells （P<0.01). Conclusion： Uric acid can promote the proliferation and migration of prostate cancer cells, which may be attributed to the down regulation of MIR22HG expression.
