Effects of ginsenoside-Rg1 on ischemia-induced angiogenesis in the mouse hindlimb

Abstract:Objective: To investigate whether Rg1 could improve angiogenesis in ischemic hindlimb of mice in vivo. Methods: Mouse hindlimb ischemia model was established by ligating the right femoral artery. Animals were randomly divided into the following two groups(n=10) for intraperitoneal treatment: Rg1(10 mg·kg-1·d-1) treated group and saline as control group. The blood flow was assessed by laser Doppler imaging at 2 weeks after surgery. Ischemic muscle was harvested for immunofluorescence analysis for CD31, TUNEL assay for apoptotic activity and colorimetric determination of nitric oxide(NO)production at 2 weeks postsurgery. Results: Rg1 promotes angiogenesis and blood reperfusion recovery. Moreover, Rg1 treatment could also significantly improve secretion of NO, and inhibit cell apoptosis in ischemic tissues. Conclusion: Rg1 promoted angiogenesis in ischemic hindlimb of mice and could be considered as a potential agent for therapeutic angiogenesis.
