Abstract:Objective: To detect the expression of CD8+CD25+Foxp3+ regulator T cells (CD8+Treg) in asthma patients, and to investigate the role of CD8+Treg in the pathogenesis of asthma patients. Methods: The study included 50 patients with asthma, 25 patients with mild-to-moderate asthma(MA), 25 patients with severe asthma(SA) and 25 age-matched healthy controls. The percentage of CD8+Treg cells was detected by flow cytometry. The Foxp3 mRNA of peripheral blood mononuclear cell was determined by RT-PCR. The cytokines of IL-33, IL-1β and transforming growth factor-β1(TGF-β1) were detected using Luminex200. Forced expiratory volume in 1 second(FEV1) and peak expiratory flow(PEF) were detected by Pulmonary Function Testing Systems. Results: Compared with healthy controls\[6.48(5.21-6.92)%], MA patients [4.13(2.05-5.41)%] and SA patients\[2.32(1.24-3.63)%] had lower percentages of CD8+Treg cells(P<0.05). Compared to healthy controls, higher levels of IL-33 were found in SA and MA patients and much higher in SA patients (P<0.05). Compared to healthy controls and MA group, SA patients showed higher levels of TGF-β1 and IL-1β. The percentage of CD8+Treg cells showed a positive correlation with FVE1(r=0.411, P=0.003) and PEF (r=0.441, P=0.001) in asthma patients. The ROC curve analysis showed the CD8+Treg had high specificity and sensitivity in the diagnosis of asthma. Conclusion: The decreased CD8+Tregs might play a role in the pathogenesis of asthma.
