Effect of Kecuan granule on Th1/Th2 cells in peripheral
blood from children with hot asthma

Abstract:Objective: To explore the effect of Kecuan granule on Th1/Th2 cells in peripheral blood from children with hot asthma. Methods: Fortyeight children with hot asthma were randomly divided into granule group and routine group, 24 cases in each group. The granule group received the administration of kecuan granule, three times per day, 1 course of 3-d treatment, for 2 courses; and the routine group received the administration of routine western medicine, pulmicort turbuhaler, 10 μg/inhaling, and terbutaline sulfate aerosol, 0.25mg/spray, twice per day, for one week. Additionally, 20 healthy children served as controls. The mRNA levels of T-box expressed in T cells (T-bet) and GATA binding-3 (GATA-3) in human peripheral blood mononuclear cells (PBMC) were detected by quantitative realtime PCR and the levels of interferon-γ(IFN-γ) and IL-4 in plasma were detected by ELISA. Results: Total effective rates of the granule group and routine group were 95.83% and 91.67%, respectively, showed no significant differences (χ2=0.559, P>0.05). Before treament, the levels of GATA3 mRNA and IL-4 in the granule group and routine group were notablely higher than those in the control group (P0.05), but the ratios of T-bet mRNA/GATA-3 mRNA and IFN-γ/IL-4 in the granule group and routine group were significantly lower than those in the control group (P0.05), but in the granule group and routine group, the levels of GATA-3 mRNA and IL-4 were markedly lower than before treatment (P0.05). Conclusion: Children with hot asthma showed Th2 accentuation and the imbalance of Th1/Th2, which could be cured by kecuan granule through inhibiting Th2 cells and correcting the imbalance of Th1/Th2.
