Abstract:Objective: To analyze the expression of microRNA in gastric cancer by bioinformatics. Methods: The differential expression of microRNAs in two subsets of GEO dataset was analyzed. Using R software package. The expressions of microRNAs in 60 gastric cancer surgical specimens and 20 paracancerous surgical specimens were detected by qRT-PCR. The relationships between microRNA expression and differentiation, TNM stage, sex, age, invasion, family history and tumor diameter were analyzed. Results: The expression of miRNA-125b-5p was up-regulated in GSE93415 and GSE99415, and the difference was statistically significant(P<0.05). The similar expression trend was confirmed in gastric cancer and normal gastric tissues with qRT-PCR. The expression of miRNA-125b-5p was correlated with TNM stage and invasion degree(P<0.05), but not with age, sex, family history, tumor diameter and differentiation degree(P>0.05).The survival time of gastric cancer patients with high expression of miR-125b-5p was significantly reduced. Conclusion: miR-125b-5p is associated with TNM stage, invasion and survival time of gastric cancer.
