Abstract:[Abstract\]Objective: To investigate the protective effects of clodronate liposome(LC) on myocardial injury by severe acute pancreatitis (SAP) in rats. Methods: A total of 48 Sprague-Dawley rats were selected into Sham group, SAP group and SAP+LC group. Each group was subdivided into 2 and 6 h groups after the operation. There were 8 rats in each group. The changes of serum IL-6, amylase and TNF-α in each group rats were measured. The effects of LC on myocardial tissue in SAP rats were observed by HE staining. The effect of LC on intercellular macrophage in rat myocardium was observed by the prussian blue staining. The dynamic changes of MAPK expression in SAP rats were observed by immunohistochemical staining. Results: The contents of IL-6, amylase and TNF-α were significantly higher at 2, 6 h after the operation in SAP+LC treatment group and SAP group than those of Sham group (P<0.05). The contents of IL-6, amylase and TNF-α in SAP+LC treatment group were significantly lower than those in SAP group (P<0.05). Compared with Sham group, the histological scores at 2, 6 h were significantly increased in SAP group and SAP+LC group(P<0.05). The histological scores at 2, 6 h in the SAP+LC group were significantly lower than SAP group(P<0.05). The number of macrophages were increased in SAP group compared with SAP+LC group at 2, 6 h by prussian blue staining. Compared with Sham group, the MAPK scores at 2, 6 h in SAP group were significantly increased(P<0.05). The MAPK scores at 2, 6 h in SAP group were significantly increased than SAP+LC group(P<0.05). Conclusion: Clodronate liposomes can reduce myocardial injury caused by SAP in rats by selectively removing monocytes/macrophages.
