Abstract:A nearinfrared region(NIR) fluorescent probe 1 was constructed with hemicyanine attached to 7nitro2,1,3benzoxadiazole (NBD) via ether bond and employed to detect the exogenous and endogenous H2S in live cells. Probe 1 is nonfluorescent due to the quenching effect of NBD. However, with the addition of H2S, the H2Slabile NBD ether bond is cleaved by thiolysis, and hemicyanine is released to lead to the enhancement of fluorescence intensity at 725 nm. In the presence of 10 equivalent of H2S, the 8.5fold fluorescence intensity enhancement is occurred. Probe 1 shows good linearity with H2S concentration when it ranges from 0.1 μmol·L-1 to 100.0 μmol·L-1 with R2 of 0.995 2. The detection limit is 0.03 μmol·L-1. Probe 1 shows NIR excitation and emission with high sensitivity, fast response and high selectivity. The constructed probe 1 can be used for monitoring and imaging exogenous and endogenous H2S in live cells.
