通过粟胁迫萌发生产高羟自由基抑制作用水溶性蛋白的方法,研究具有羟自由基抑制作用粟水溶性蛋白的胁迫培养基.试验表明,H2O2和复合蛋白酶对萌发粟水提取物的羟自由基抑制作用起关键作用.运用析因试验、中心组合试验和响应面分析方法筛选、优化,得到粟胁迫萌发培养基为:体积分数1.56%的H2O2,0.259 6 mg/L赤霉素、0.248 g/L复合蛋白酶、体积分数0.7%的吐温-40.使用此培养基胁迫萌发,粟的水溶性蛋白羟自由基抑制作用达58.16%.
Abstract
A production method of high hydroxyl radical inhibition by water-soluble protein from stress millet was developed. The test indicated that H2O2 and Protamex play key roles in the hydroxyl radical inhibitory activity. Fractional factorial design, central composite experimental design and response surface methodology were used to optimize those factors. The optimal stress medium composition was listed as follows : H2O2 1.56 % , gibberellin 0. 259 6 mg/L, Protamex 0. 248 g/L and Tween -40 0.7%. With the optimal medium, the highest hydroxyl radical inhibition (58.16%) was achieved.
