Inhibitory effect of the ginsenoside Rb-2 on replication of enterovirus 71 in vitro

Abstract Objective: To investigate the inhibitory effect of ginsenosides Rb-2 on enterovirus 71(EV71). Methods: The cytotoxicity of ginsenosides Rb-2 on human embryonic rhabdomyosarcoma(RD) cells was detected by MTT method.The inhibitory effect of ginsenosides Rb-2 on EV71 was observed by cytopathic effect method, and the influence of the drug on direct inactivating and absorbing effect was also studied. Results: When ginsenosides Rb-2 was at a concentration of ≤100 μg/mL,there was no significant cytotoxicity on RD cells. At 6.25, 12.5, 25, 50, 75 and 100 μg/mL, the inhibitory rates of ginsenosides Rb-2 against EV71 were (16.0±2.8)%,(23.4±2.5)%,(61.4±3.1)%, (77.9±2.9)%,(98.3±0.8)% and (98.6±1.0)%,respectively.The 50% inhibitory concentration was 20.8 μg/mL.Treated by the mixture of ginsenosides Rb-2 (≤100μg/mL) and EV71,the direct inactivating effect on EV71 wasn′t found in RD cells. When both ginsenosides Rb-2(75 μg/mL)and EV71 were added, or added at 1 hr postinfection, there was no statistical significance in viral titers. Conclusion: Ginsenosides Rb-2 can effectively inhibit EV71 replication.
