Effect of type Ⅱ cGMP-dependent protein kinase on tumor suppressors in breast cancer cell line MCF-7 |

Abstract Objective: To investigate the effects of PKGⅡ on tumor suppressor genes including PTEN,p21,p53,breast cancer type 1 susceptibility protein (BRCA1) and BRCA2 in breast cancer cell line MCF-7. Methods: MCF-7 cells were transfected with adenoviral vector encoding cDNA of PKGⅡ and the kinases were activated by respective activator. Western blotting was applied to detect the influence of PKGⅡ on the expression of tumor suppressor proteins, including p53,p21,PTEN, BRCA1 and BRCA2. Results: When PKGⅡ expression and activity were increased, the expression of PTEN,p21,p53,BRCA1 and BRCA2 was decreased. Conclusion: PKGⅡ could inhibit the expression of PTEN,p21,p53,BRCA1 and BRCA2.
