miR-129 regulates epithelial-mesenchymal transition in lung adenocarcinoma cell A549 via downregulating TCF4 expression |

Abstract Objective: To investigate the regulatory effects and mechanism of miR129 on TCF4 expression and epithelialmesenchymal transition(EMT) in lung adenocarcinoma cell A549. Methods: miR129 mimic was transiently transfected into lung adenocarcinoma cell A549.The expression levels of miR129 were determined by qRTPCR.The differential expression of TCF4, Ecadherin, vimentin were detected by qRTPCR and Western blotting. Reporter gene plasmid including TCF4 3′UTR was constructed and miR129 oligonucleotides (miR129 mimic) was also synthetized. miR129 mimic and reporter gene plasmid were cotransfected into HEK293 cells, and then luciferase activity of TCF4 3′UTR report gene was detected. Results: miR129 mimic decreased the expression of TCF4 and vimentin, and increased Ecadherin expression(P<0.05 or P<0.01). Luciferase reporter assay indicated inhibition of luciferase activity of TCF4 and TCF4 3′UTR by miR129(P<0.01). Conclusion: miR129 suppressed TCF4 expression and activation ofEMT signal pathway in lung adenocarcinoma cell A549.
