The expression of calciumsensing receptor (CaSR) and the effect of CaSR
inhibitor NPS2390 after spinal cord ischemiareperfusion injury in rats |

Abstract Objective: To investigate the expression change of calciumsensing receptor(CaSR) and the effect of CaSR inhibitor NPS2390 on spinal cord ischemiareperfusion injury in rats. Methods: Fortyeight SD rats were randomly divided into sham operation group(n=16), model group (n=16) and NPS2390 group (n=16). The sham operation group only opened the abdomen, did not clamp the abdominal aorta, the model group blocked the abdominal aorta after the reperfusion, the NPS2390 group before premodeling 2 d received injection of CaSR inhibitor NPS2390 through caudal vein. Each group was divided into 4 subgroups according to postoperation 6 h, 12 h, 24 h and 48 h. Motor function of hindlimb of rats were measured by BBB score. CaSR expression were measured by Western blotting and immunohistochemical staining, and the apoptosis of spinal cord tissue cells was detected by TUNEL staining. Results: The BBB score of model group and NPS2390 group was significantly lower than sham operation group(P0.05). CaSR expression in model group was significantly increased at each time point(P0.05). Conclusion: The expression of CaSR increased gradually and peaked at 24 h after spinal cord ischemia reperfusion injury in rats, and CaSR inhibitor NPS2390 exerted protection against spinal cord ischemia reperfusion injury in rats.
