Effect of HMGB1 on the expression of TGF-β1 of cadiac fibroblast induced by high glucose |

Abstract Objective: To investigate the effect of HMGB1 on the expression of TGF-β1 of cadiac fibroblast induced by high glucose.Methods: The cardiac fibroblasts were isolated from 3 w C57/BL6 mice.The identification of cardiac fibroblasts was evaluated by immunocytochemical staining with vimentin. The cardiac fibroblasts were treated by high glucose for different times(0,6,12,24,48 h).After transfected with siRNA, the cardiac fibroblasts were treated by high glucose for 6 h. The mRNA levels of HMGB1 and TGF-β1 of cardiac fibroblasts were determinated by RT-PCR.Results: The cultured cells were in accordance with characteristics of cardiac fibrolast. The mRNA levels of TGF-β1 of cardiac fibroblasts were inhibited by HMGB1siRNA after treated by high glucose for 6 h. Conclusion: HMGB1 may contribute to modulate the expression of TGF-β1 of cardiac fibroblasts induced by high glucose.
