Abstract:Objective: To investigate protective effect of lycopene on liver fibrosis in mice induced by concanavalin A (Con A). Methods: Twentyfour mice were equally randomized into 3 groups: control group, model group and prevention group. The mice in model group was received intraperitoneal injection of CoA(20 mg/kg),one time per week, while control group received normal saline, prevention group was received intragastric administration of lycopene, one time per day, for 8 weeks. Mice blood were collected to test the indices of hepatic fibrosis. Grade of hepatic fibrosis were evaluated according to the semi-quantitative scoring system (SSS). Western blotting was used to test the expression of α-smooth muscle actin(α-SMA), Smad3 protein(Smad3) and transforming growth factor-β1 (TGF-β1). Results: Compared with the control group,the model group showed higher serum levels of hyaluronic acid (HA), laminin (LN), procollagen type Ⅲ(PCⅢ) , SSS score, malonaldehyde level, the expression of α-SMA, TGF-β1 and Smad3, lower superoxide dismutase (SOD) (both P<0.01); compared with the model group, the prevention group showed lower serum levels of HA, LN, PCⅢ, SSS score, malonaldehyde level, the expression of α-SMA, TGF-β1 and Smad3, higher levels of SOD (both P<0.01). Conclusion: Lycopene could sharply decrease the harm of peroxidation, protect the liver cells, reduce the collagen production and restrain the expression of the important factor of α-SMA, Smad3, TGF-β1 which lead to liver fibrosis, so that lycopene had the notable alleviate effect on liver fibrosis induced by ConA.
