Objective: To investigate the effect of Survivin overexpression on the expression of apolipoprotein E (ApoE) in 3T3-L1 adipocytes.Methods: The lentiviral expression vector for Survivin and its control vector were constructed and transfected into 3T3-L1 preadipocytes. Then the 3T3L1 adipocytes were cultured and differentiated, and transcriptome analysis was performed on the eighth day of differentiation. The mRNA level of ApoE in 3T3-L1 adipocytes was determined by qRT-PCR. Intracellular and secreted ApoE protein levels were measured by Western blotting. Results: The 3T3-L1 adipocyte cell line with stable overexpression of Survivin was successfully constructed.Overexpression of Survivin does not affect the differentiation of adipocytes. Transcriptome analysis showed high expression of ApoE in basal condition. Among the apolipoprotein family members, ApoE is down-regulated specifically by Survivin overexpression. The mRNA and protein levels of ApoE in the Survivin overexpression group were significantly lower than those in the control group(P<0.05 and P<0.01). The expression level of secreted APOE in the high expression group was also significantly lower than that in the control group(P<0.01). Conclusion: High expression of Survivin in 3T3-L1 adipocytes can inhibit the expression of ApoE at the transcriptional and protein levels.
