Abstract:Objective: To investigate the effect of atractylodin on proliferation of human colon cancer LS174T cells. Methods: LS174T cells were divided into six groups, including blank control group, absolute ethyl alcohol control group, and atractylodin-treated groups(10, 20, 40, 80 mg/L). The changes of cell morphology were observed with optical microscope; inhibition of cell proliferation by atractylodin was measured with MTT assay; cell cycle analysis was analyzed with flow cytometry; the cytokine expression was measured with liquichip method. Results: Atractylodin induced LS174T cell growth inhibtion in a time- and concentration-dependent manner. G0/G1phase ratio of LS174T cells treated by atractylodin was increased. Atractylodin inhibited the expression of IL-6 and IL-7 in LS174T cells. Conclusion: Atractylodin could inhibit the proliferation of LS174T cells in a dose- and time-dependent manner, which may be caused by blocking the cell cycle progression and the expression of associated cytokines.
