93 Clinical significance of platelet parameters in preterm infants with bronchopulmonary dysplasia

Objective:  To study the dynamic changes of platelet parameters in preterm infants with bronchopulmonary dysplasia (BPD) and its clinical significance. Methods: From January 2015 to March 2021, preterm infants with gestational age less than 32 weeks were admitted to Affiliated Hospital of Jiangsu University, whose clinical data and platelet parameters such as platelet count, mean platelet volume, platelet distribution width and platelet mass index were retrospectively analyzed, and the clinical data and platelet parameters of each group were also compared. Logistic regression was used to analyze the relationship between platelet parameters and BPD, and receiver operating characteristic curve was used to analyze the predictive value of platelet count and platelet mass index for BPD. Results: A total of 224 premature infants, 48 with BPD and 176 with nonBPD were included. The platelet count and platelet mass index of BPD on first day were lower than those of nonBPD. After adjusting for gestational age, birth body weight, patent ductus arteriosus and the use of pulmonary surfactant, platelet mass index (OR=0.91, 95% CI: 0.850-0.945) and platelet count (OR=0.96, 95% CI: 0.924-0.986) were independent risk factors for BPD. Platelet mass index (AUC=0.766, 95% CI: 0.681~0.855, P<0.01) and platelet count (AUC=0.668, 95% CI: 0.566~0.770, P<0.01) could predict the occurrence of BPD. Conclusion: The low platelet mass index and platelet count in preterm infants may be related to the occurrence of BPD. Postnatal platelet mass index and platelet count may be used for predicting the occurrence of BPD.

99 Bioinformatical analysis of key genes and pathways of bronchopulmonary dysplasia based on GEO

Objective:To explore the key genes and signal pathways of bronchopulmonary dysplasia (BPD) using bioinformatics. Methods:  Data sets were screened in the gene expression omnibus (GEO) database, the GEO2R tool was used to screen the differentially expressed genes between BPD and extremely preterm infants without BPD. Functional annotation and pathway analysis were performed using the DAVID database, proteinprotein interaction (PPI) network construction of key DEGs was completed using the STRING online tool. The core genes were screened and visualized by Cytoscape software. Results: A total of 375 genes were up-regulated and 390 genes were downregulated via differential analysis. GO enrichment analysis showed that differentially expressed genes were involved in cellular response to calcium ions responsiveness, transcriptional regulation of RNA polymerase Ⅱ promoter, and heart development. KEGG enrichment analysis showed that differentially expressed genes were concentrated in glucagon signaling pathway and breast cancer signaling pathway. Ten core genes were identified from the constructed PPI network, namely JUN, FOS, SMARCA4, ATF3, EGR1, ERBB2, PPARA, FOSB, NR4A1 and IGF1. Conclusion:  JUN, FOS, ATF3 and EGR1 may play an important role in the pathogenesis and development of BPD.

113 Expression of BRCA1, BRCA2, RAD51 and FANCD2 in ovarian cancer tissues and correlation with clinicopathological characteristics

Objective: To explore the protein expression of homologous recombination repair genes for DNA doublestrand breaks, such as breast cancer susceptibility protein 1 (BRCA1), breast cancer susceptibility protein 2 (BRCA2), DNA repair protein RAD51 and Fanconi anemia D2 protein (FANCD2), in ovarian cancer tissues and correlation with clinicopathological characteristics. Methods: Paraffin sections of 84 cases of primary epithelial ovarian cancer (EOC) and 6 cases of normal ovarian tissues were collected, and the levels of protein expression of BRCA1, BRCA2, RAD51, and FANCD2 were detected by immunohistochemistry. The expression of the above four proteins in the EOC tissues and the correlation between the proteins in the cancer genome atlas (TCGA) database through GEPIA. Ualcan analyzed the relationship between the expression of the four proteins and the clinicopathological characteristics of EOC in the TCGA database. Results: Immunohistochemical assays revealed that BRCA2, FANCD2 and RAD51 were highly expressed in EOC specimens rather than in normal tissues (all P<0.05). According to the analyses of TCGA database by GEPIA, the mRNA expression of BRCA1, BRCA2, RAD51 and FANCD2 increased in EOC tissues. The expression levels of 4 proteins have a significant positive correlation between each other. According to the results of TCGA database by Ualcan analysis, the mRNA expression of BRCA1 was significantly higher in patients over 41 years of age; expression of these four genes were higher in tumors above grade 2 and the mRNA expression of RAD51 in grade 3 tumors was significantly higher than that in grade 2 tumors; the mRNA expression of FANCD2 in African American was significantly higher than that in Caucasian; the mRNA expression of BRCA1, RAD51, and FANCD2 in TP53mutant tumors was significantly higher than their expression in nonmutant tumors. Conclusion: The expression of BRCA2, FANCD2 and RAD51 was significantly increased in tumor tissues of EOC, and may potentially be candidate biomarkers for diagnosis and treatment of ovarian cancer.

118 The expression and clinical significance of miR-30a and related

[Abstract]Objective: To investigate the expression of miR-30a and related inflammatory indicators in sepsis and analyze its clinical significance based on bioinformatics. Methods: The gene chips were searched and screened by GEO database and their expression was analyzed. The target genes were predicted by the prediction software of miR-30a website database and KEGG pathway enrichment analysis was performed. A total of 64 sepsis patients admitted to Affiliated Hospital of Jiangsu University from January 2019 to January 2021 were collected and divided into non-shock group (n=32) and shock group (n=32) according to their severity. According to the outcome of 28 d, the sepsis patients were divided into survival group (n=44) and death group (n=20). Ten healthy subjects were selected as control group. The levels of white blood cells, C-reactive protein, procalcitonin and interleukin-6 in each group were detected, and sequential organ failure score (SOFA) were calculated. The expression of miR-30a in peripheral blood was detected by RT-PCR, and the correlation between miR30a and SOFA score was analyzed. ROC curve was used to evaluate the diagnostic value of miR-30a in sepsis. Results:   GEO database retrieval showed that miR30a expression in sepsis group was higher than that in control group (P<0.05). Compared with the control group, miR-30a, white blood cells, C-reactive protein, procalcitonin, interleukin-6 and SOFA scores were increased in the sepsis group(P<0.05 or <0.01). The white blood cells and SOFA scores in the shock group were higher than those in the nonshock group (P<0.05). Compared with sepsis survival group, SOFA score was higher in death group(P<0.05). Spearman correlation analysis showed that the relative expression of miR30a was positively correlated with SOFA score of sepsis (r=0.751, P<0.01) and the expressions of interleukin-6, C-reactive protein and procalcitonin (r=0.667, 0.736, 0.496, P<0.01). ROC curve evaluation results showed that the area under the curve of miR-30a was 0.831(P<0.01), the sensitivity was 89.1%, and the specificity was 80.0%; the area under the curve of C-reactive protein was 0.841(P<0.01), the sensitivity was 79.7%, the specificity was 90.0%. The area under the curve of miR30a+C-reactive protein was 0.902(P<0.01), the sensitivity was 89.1%, and the specificity was 90.0%. Conclusion: miR30a and related inflammatory markers are highly expressed in sepsis, which may be potential markers for the diagnosis of sepsis.

124 Expression of SOX7 in renal cell carcinoma and its effect on the biological behavior of renal cancer cells

Objective: To investigate the expression of sex determining region Y-box 7(SOX7) in renal cell carcinoma and the effect on the biological behavior of renal cancer cells. Methods:  qRT-PCR and methylationspecific PCR (MSP) were used to detect the expression and methylation status of SOX7 in renal cell carcinoma tissues and renal cancer cells (A498, ACHN, 786O); immunohistochemistry (IHC) was used to detect the protein expression of SOX7 in renal cell carcinoma tissues. The SOX7 mRNA expression in renal cancer cells after 5-Aza-dC/TSA intervention was also detected by qRT-PCR. After transfection of SOX7 overexpression plasmid in renal cancer cells ACHN, cell proliferation, migration and invasion were detected by MTS, clone formation, scratch assay and Transwell assay, respectively. Results:  The qRT-PCR and IHC results showed that the expression of SOX7 mRNA and protein in renal cancer tissues was significantly lower than that in paraneoplastic tissues (P<0.01) and correlated with lymph node metastasis and TNM grade (P<0.05). SOX7 mRNA expression in three renal cancer cells (A498, ACHN, 786O) was significantly lower than that in the control group HK-2 (P<0.01). SOX7 mRNA expression was increased after 5-Aza-dC/TSA treatment in all (P<0.01). MSP results showed that the methylation rate of SOX7 was significantly higher in kidney cancer tissues than that in paraneoplastic tissues (P<0.01) and correlated with TNM stage (P<0.05). Overexpression of SOX7 inhibited the proliferative activity, migration ability and invasive ability of ACHN. Conclusion:  SOX7 is lowly expressed in renal cell carcinoma, hypermethylation of SOX7 may be one of the molecular mechanisms of renal carcinoma histogenesis.

131 Effect of transcription factor GATA binding protein 3 on the  expression of asthma susceptibility gene ADAM33

Objective: To investigate the effect of transcription factor GATA binding protein 3(GATA3) on promoter activity and mRNA expression of a disintegrin and metalloproteinase 33(ADAM33). Methods:  The 1 953 bp fragment of ADAM33 promoter region was amplified by PCR based on human normal lung epithelial BEAS-2B cell DNA as template. The recombinant plasmid of ADAM33 promoter was cloned into pGL3-Basic vector by double digestion, ligation and transformation, and then transfected into HEK293T and BEAS-2B cells. The promoter activity of the constructed plasmid of ADAM33 gene in HEK293T and BEAS-2B cells were detected by double luciferase reporter gene technology. The HEK293T and BEAS-2B cells were divided into pcDNA-GATA3 group and pcDNA3-1 control group, respectively, and transfected with GATA3 overexpression plasmid and blank control plasmid,respectively; the luciferase activity was detected by dual luciferase reporter gene technology; the mRNA expression of ADAM33 in BEAS-2B cell was detected by real-time fluorescence quantitative PCR. Results:  The recombinant plasmid containing ADAM33 promoter fragment was successfully constructed by PCR, double enzyme digestion and sequencing. Compared with the pcDNA31group, the luciferase activity in the ADAM33 promoter region of pcDNA-GATA3 group in HEK293T and BEAS2B cells was significantly increased(P<0.05 or P<0.01); the pcDNAGATA3 group showed higher level of the relative expression of ADAM33 mRNA(P<0.01). Conclusion:  Transcription factor GATA3 up-regulates the promoter activity and mRNA expression of human asthma susceptibility gene ADAM33.

136 Inhibitory effect of semi-compatible cell transplantation on CT-26 colon cancer cells in tumorbearing mice

Objective: To investigate the effect of grafts transplanted with haploidentical cells on CT-26 colon cancer cells in tumor-bearing mice and its potential mechanism. Methods: The spleen cells and bone marrow cells of C57BL/6 mice were taken as donor cells; 18 CB6F1 mice(receptor) were taken and randomly divided into control group, cyclophosphamide group and transplantation group, 6 mice in each group, and subcutaneously implanted mouse colon cancer CT26 cells to establish subcutaneous tumorbearing mouse model; then 0.2 mL of normal saline, cyclophosphamide, cyclophosphamide+spleen cells+bone marrow cells were injected into the tail vein for 4 consecutive weeks; the tumor volumes of the three groups mice were detected at different time points; tumor, liver, small intestine and skin tissues of three groups mice were harvested at 2 weeks, and the pathological changes were observed by HE staining; contents of IL2, IL-4, IL-10, IFN-γ, TNF-α and TGF-β in mouse tumor tissues were detected by ELISA. The chimerism rate of peripheral blood cells (H-2Kd/b) in the transplantation group at 3 days, 2 weeks and 4 weeks was detected by flow cytometry. Results: At the 2-week, the tumor volume in the cyclophosphamide group and the transplantation group was significantly smaller than that in the control group(both P<0.01). Compared with the control group and cyclophosphamide group, the contents of IL-2, IL-10, IFN-γ and TNF-α in tumor tissues of the transplantation group were significantly increased(all P<0.05), while the contents of IL-4 and TGF-β were significantly decreased(both P<0.05). Compared with those at 3-day, the proportion of H-2Kb in peripheral blood of CB6F1 mice was significantly increased at 2- and 4-week(both P<0.01). Conclusion:  Splenocytes and bone marrow cells derived from C57BL/6(H2Kb) mice exerted an anticolorectal cancer effect on tumorbearing mice by changing the immune microenvironment of CB6F1(H-2Kd/b) mice.

141 Correlation of speed of blastocyst development and morphological parameters with embryo euploidy and pregnancy outcomes

Objective: To investigate the correlation among the blastocyst development speed, the degree of blastocyst expansion, the level of inner cell mass, and the level of trophoblast cells and their euploidy status and pregnancy outcomes in preimplantation genetic testing cycles. Methods: A total of 237 preimplantation genetic testing for aneuploidy(PGT-A) cycles and 127 resuscitation transplant cycles from October 2017 to January 2021 in the Center of Reproductive Medicine, Obstetrics and Gynecology Hospital Affiliated to Nanjing Medical University were included in the study. The relationship among embryo development speed and morphological characteristics, euploidy and pregnancy outcome was analyzed. Results: ① In 237 biopsy cycles included for analysis, 975 blastocysts were successfully detected, including 479 euploid blastocysts(49.13%) and 496 aneuploid blastocysts(50.87%).② After considering the correction factors, the euploid rate of blastocysts on the 6th and 7th day was significantly lower than that on the 5th day(OR=0.640, 95% CI: 0.493~0.830, P=0.001). The euploidy rate of cell masses in blastocyst grade B was significantly lower than that in grade A (OR=0.235, 95% CI: 0.123-0.449, P<0.001); the euploidy rate of blastocyst trophoblast of grade C was significantly lower than that of biopsy blastocyst of grade A (OR=0.360, 95% CI: 0.192-0.674, P=0.001). The degree of blastocyst expansion has no association with the euploid state. ③ Before and after age correction, there were no difference in clinical pregnancy rate between blastocyst formation time group, blastocyst expansion degree group, inner cell grade group and trophoblast cell group. Conclusion: In PGT-A cycles, the blastocyst development rate, the grade of inner cell mass and the grade of trophoblast cells were significantly correlated with the status of euploidy, while the degree of blastocyst expansion was not correlated with the status of euploidy. There was no correlation between the early pregnancy outcomes of euploidy embryo transfer cycles and the time of blastocyst formation, the grade of inner cell mass, the grade of trophoblast cells and the degree of blastocyst expansion.

146 Neutrophil chemotaxis dysfunction and potential mechanisms in diabetes mellitus#br#

Objective: To investigate whether the chemotactic function of neutrophils in type 2 diabetes is impaired and its underlying mechanism. Methods: According to the WHO diagnostic criteria, a total of 45 patients with diagnosed type 2 diabetes and 45 healthy controls of the same age were recruited. The tested neutrophils were all collected from the peripheral blood of the subjects′ cubital veins. The chemotactic function of neutrophils in the two groups was detected by the modified chemotaxis method under agarose, and the expression level of P2X1 receptor on the surface of neutrophil membrane was detected by flow cytometry and immunofluorescence. In the intervention experiments, the normal neutrophils were stimulated by diluted plasma of patients with type 2 diabetes or healthy controls (90%, 20%), media with different glucose contents (5, 12 and 25 mmol/L), end products of glycation (0.5,1, 2 and 4 mmol/L) and palmitic acid (50, 100 and 200 μmol/L) and the chemotaxis were detected. Results: Compared with healthy controls, the chemotactic function of neutrophils type 2 diabetes was significantly reduced (P<0.001), and the expression of P2X1 receptors increased (P<0.001). There was a significantly negative correlation between them (r=-0.60, P<0.001). Compared with the control group, the patients′ highconcentration plasma significantly inhibited the chemotactic function of neutrophils (P<0.001). Each concentration of glycation end products and glucose was not significantly inhibited neutrophils chemotaxis function (P>0.05). While high concentrations of palmitic acid (200 μmol/L) significantly inhibited the chemotaxis of neutrophils (P<0.05), the expression of P2X1 receptors on the cell surface increased (P<0.05), and there was a significantly negative correlation (r=-0.67, P<0.05) between them. Conclusion: The chemotaxis of neutrophils is impaired in type 2 diabetes, and high concentrations of palmitic acid impairs the chemotaxis of neutrophils through P2X1 receptors.

152 Learning curve of minimally invasive total hip arthroplasty with SuperPATH approach#br#

Objective: To investigate the clinical efficacy and learning curve of minimally invasive total hip arthroplasty (THA) with supercapsalar percutaneously assisted total hip(SuperPATH) approach.Methods: A total of 102 patients with hip disease treated by SuperPATH approach, the surgery situation, postoperative complications, acetabular abduction and anteversion, postoperative 24 h visual analogue scale(VAS) score and Harris hip joint function score on day 7 were recorded. The learning curve of operation was explored and the above indexes were comparatively analyzed.Results: All patients were followed up for 6-25 months.Operation time, intraoperative blood loss,and hospital stay tended to be stable after the completion of 36 cases of SuperPATH-THA. The 102 cases were divided into the learning period (36 hips) and the stable period (66 hips). In the stable period, the operative time, intraoperative blood loss, the first time to the ground, hospital stay were significantly reduced compared with those in the learning period (all P<0.01). There was no significant difference in the length of the incision, acetabular abduction and anteversion, postoperative 24 h VAS score and Harris score after surgery between the two periods (all P>0.05). One case of prosthetic dislocation and 1 case of periacetabular fracture in the learning period.Conclusion: SuperPATH approach minimally invasive THA is consistent with the concept of minimally invasive and rapid rehabilitation, has the advantages of shorter hospital stay, and lower complication rate; the learning curve of SuperPATHTHA was about 36 cases;after passing the learning curve, the residents could provide better early postoperative outcomes for patients.

156 Clinical efficacy of levocarnitine combined with Jinshuibao in the treatment of patients with chronic renal failure undergoing maintenance hemodialysis#br#

Objective: To evaluate the clinical effect of levocarnitine combined with Jinshuibao on patients with chronic renal failure patients undergoing maintenance hemodialysis. Methods:  Ninetytwo patients with chronic renal failure who underwent maintenance hemodialysis in Yongchuan Hospital Affiliated to Chongqing Medical University from June 2018 to February 2021 were selected in the study, and were randomly divided into the treatment group (n=46) and the control group (n=46). The control group was treated with levocarnitine, and the treatment group was treated with Jinshuibao on the basis of the control group. Clinical efficacy and level of inflammatory factors, matrix metalloproteinases, nutritional indicators, stromal cell-derived factor-1 (SDF-1), vitamin D and vitamin B12 in serum were compared between the two groups. Results:After treatment, the total clinical effective rate of treatment group (89.13%) was significantly higher than that of control group (69.57%, P<0.05). After treatment, the levels of tumor necrosis factor-α(TNF-α), IL-6, C-reactive protein (CRP), matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-9 (MMP-9) and SDF-1 in 2 groups were significantly decreased compared with before treatment(all P<0.05). The levels of total protein, albumin, hemoglobin, vitamin D and vitamin B12 were higher than those before treatment(all P<0.05); and the contents of TNF-α, IL-6, CRP, MMP-2, MMP-9 and SDF-1 in treatment group were lower than those in control group (P<0.05), and the levels of total protein, albumin, hemoglobin, vitamin D and vitamin B12 in treatment group were higher than those in control group(P<0.05). There was no significant difference in the incidence of adverse reactions and complication between the two groups(both P>0.05). Conclusion:  Levocarnitine combined with Jinshuibao could improve the clinical efficacy, reduce microinflammatory reaction, improve nutritional status in patients with chronic renal failure undergoing maintenance hemodialysis.

161 Effect of chitosan combined with nano-silver on wound recovery in patients with high-grade squamous intraepithelial lesion after LEEP treatment#br#

Objective: To investigate the effect of chitosan combined with nanosilver on the wound recovery of patients with highgrade squamous intraepithelial lesion (HSIL) who underwent loop electrosurgical excision procedure (LEEP) treatment. Methods: A total of 160 patients who were admitted to the Fourth Affiliated Hospital of Jiangsu University from September 2020 to May 2021 pathologically diagnosed with HSIL and negative margins after LEEP treatment were enrolled in the study. Then the patients were randomly divided into control group, chitosan group, nanosilver group, combined group (chitosan combined nanosilver) and control group, with 40 cases in each group. Except for the control group, the other three groups were administrated with the corresponding treatment twice a week, for a total of 8 times. In each group within 2 months after the operation, the wounds after the operation, the cleanliness of the vaginal secretions in 2 weeks after the operation and postoperative complications were observed and recorded. ELISA was used to detect the content of inflammatory factors such as TNF-α, IL-6, IL-8 and highmobility group box 1 protein (HMGB1) in cervical secretions before and 1, 2, 3, 4 weeks after operation. Results: Compared with control group, the disappearance time of vaginal discharge, the time to scab off the wound, and the time of wound healing in the combined group were significantly reduced in chitosan group and nano-silver group(all P<0.05). Compared with the control group and the chitosan group, the incidence of lower abdominal pain and purulent secretions after operation in the combined group were greatly reduced(all P<0.05). Compared with the control group and the chitosan group, the content of HMGB1, TNF-α, IL-6 and IL-8 in combined group were significantly reduced in 1, 2, 3, 4 weeks after operation(all P<0.05). Compared with the nanosilver group, the content of HMGB1(1, 2, 4 weeks after operation), TNF-α (1, 2, 3, 4 weeks after operation), IL-6 (1, 2, 3 weeks after operation), IL-8(1, 3, 4 weeks after operation) in the combined group were remarkably decreased(all P<0.05). Conclusion: The combined application of chitosan and nanosilver could not only promote the healing of cervical wounds after LEEP treatment, but also prevent postoperative complications.

167 comValue of multimodal magnetic resonance imaging in differentiating Warthin tumor from pleomorphic adenoma of parotid gland#br#

Objective: To explore the value of multimodal magnetic resonance imaging (MRI) technology in distinguishing parotid Warthin tumor and pleomorphic adenoma. Methods: The data of patients with pathologically confirmed Warthin tumors of the parotid gland (62 cases, 66 lesions) and pleomorphic adenomas (47 cases, 49 lesions) admitted to the hospital from October 2017 to May 2021 retrospectively analyzed, all patients underwent routine MRI, diffusion weighted imaging (DWI), and dynamic contrast enhancement (DCE) examinations before surgery.Clinical characteristics, apparent diffusion coefficient (ADC) values and DCEMRI parameter values of patients with Warthin tumor and pleomorphic adenoma of the parotid gland were compared. In addition, the receiver operating characteristic (ROC) curve was used to analyze the differential value of ADC value and DCEMRI parameter value in parotid Warthin tumor and pleomorphic adenoma. Results: There were no significant difference in maximum crosssectional area of the lesion, lesion location, lesion shape, and lesion boundary between patients with Warthin tumor of the parotid gland and pleomorphic adenoma (P>0.05), the difference in the ratio of male to female in the two groups was statistically significant (P<0.001), patients with Warthin tumor of the parotid gland were less cystic than those with pleomorphic adenoma (P<0.001). The ADC value of patients with Warthin tumor of the parotid gland was lower than that of patients with pleomorphic adenoma (P<0.001). There were no statistically significant difference in the rate constant (Kep) and initial area under the contrast agent concentration curve (iAUC) between patients with Warthin tumor of the parotid gland and pleomorphic adenoma (P>0.05). The volume transfer constant (Ktrans) value of patients with Warthin tumor of the parotid gland was higher than that of patients with pleomorphic adenoma (P<0.001), the extravascular extracellular space volume ratio (Ve) of patients with Warthin tumor of the parotid gland was lower than that of patients with pleomorphic adenoma (P<0.001). ROC analysis shows that the best cutoff points for ADC, Ktrans, and Ve were 1.00×10-3 mm2/s, 0.19 min-1, 0.42, the sensitivity were 81.82%, 83.33%, 84.85%, the specificity were 73.47%, 77.55%, 69.39%, and the area under the curve (AUC) were 0.814, 0.741, 0.760, respectively. The specificity and AUC of the combination of the three were 95.92% and 0.866, respectively. Conclusion: Multimodal MRI technology has high clinical application value for the differential diagnosis of parotid Warthin tumor from pleomorphic adenoma.
