277 Relationship between increased nuocytes and GITRGITRL expression levels in lung tissue of asthmatic mice

Objective: To detect the changes of numbers of nuocytes and levels of GITRGITRL in the lung of asthmatic mice, analyze the relationship between them, then to explore the changes of Th2polarized microenvironment at the locus of allergic inflammation when with asthma. Methods: Eight mice were devided into 2 groups randomly, asthma models were sensitized with OVA on 1st, 11st day, then nebulized inhalation with OVA on day 22-26, control group with PBS. Flow cytometry was used to detect numbers of nuocytes and cells which express GITR; qRTPCR was also used to test the mRNA levels of GITRGITRL, molecules and cytokines associated with nuocytes; pearson correlation analysis was used to detect the correlation between GITRGITRL and nuocytes. Results: Compared with the control group, model group showed high levels of nuocytes and GITRGITRL, meanwhile, with increased mRNA levels of GITRL, Foxp3, RORα, ICOS, ST2, IL5 and IL13; and there was a positive correlation between GITR expression and RORα, ST2, IL5, IL13 expression. Conclusion: Nuocytes and the expression of GITRGITRL increased synchronously in lung of asthmatic mice, which suggested that the upregulation of T cellmediated immune response by GITRGITRL may be associated with Th2biased response and polarization of nuocytes.

282 Dynamic study of magnetic resonance spectrum about sensitive brain structure of rat obsessive compulsive disorder model

Objective: To analyze the metabolism of the hippocampus and frontal lobe tissue of the rat obsessive compulsive disorder model by 1HMRS. Methods: A total of 40 SD rats were randomly divided into control group and quinpirole group,10 rats in control group, 30 rats in quinpirole group. The rats in quinpirole group received quinpirole subcutaneous injection twice a week, the control group received the same amount of saline injection. All of the rats were injected ten times. Every time after delivery, all of the rats received 3.0T MRI and analyzing technology 1HMRS(indexes including NAA, Cr, Cho and MI). Results: ① Compared with the control group, the values of NAA/Cr of the bilateral hippocampus of the quinpirole group were significantly reduced from the 10th day (P<0.05), and the values began to significantly rise from the 24th day (P<0.05); the values of NAA/Cr of the bilateral frontal lobe of the quinpirole group were significantly rose from the 10th day(P<0.05), and keep the same level on the 21-34th day. ② The values of Cho/Cr of the bilateral frontal lobe of the quinpirole group were significantly rose from the 21th day (P<0.05); but the values of hippocampus had no obvious change (P>0.05). ③ The values MI/Cr of the bilateral hippocampus of the quinpirole group were significantly reduced from the 7th day, and keep the same level on the 21-34th day (P<0.05); but the values of frontal lobe had no obvious change(P>0.05). Conclusion: Many metabolite indexes were abnormal in quinpirole rat model, and some showed certain regularity.

286 Investigating the relationship between MicroRNA-195 and Smad7

Objective: The luciferase reporter vector containing 3′UTR of Smad7 was constructed. Dual luciferase reporter gene system was applied to determine the association between miRNA195 and its target gene Smad7. Methods: The 3′UTR of Smad7 fragment amplified by PCR was cloned into luciferase vector. MiRNA195 and the luciferase reporters containing 3′UTR(Smad73′UTRWT) or mutant 3′UTR(Smad73′UTRMut) of Smad7 were cotransfected into HEK293T cells and dual luciferase reporter gene system was applied to test luciferase activity. Results: DNA sequencing showed that the sequences of the cloned regions were correct.The luciferase activity of Smad73′UTRWT plasmid treated with miRNA195 was decreased to about 40% compared with control. The mutant 3′UTR were no longer repressed by miRNA195. Conclusion: The Smad7 3′-UTR luciferase reporter vector has been constructed successfully.MiRNA195 can repress the luciferase activity of the reporter gene and had direct effect on Smad7 3′-UTR.

290 Effect of interaction between BTLA and HVEM on T cell activation

Objective: To investigate the effect of interaction between B and T lymphocyte attenuator (BTLA) and HVEM of mouse bone marrowderived dendritic cells(BMDC)on T cell activation. Methods: T cells and BMDC from C57BL/6 mouse were mixed and cultured. After blocking HVEM on BMDC by antibodies against HVEM, MTT method was used to examine T cell proliferation and the secretion of IL2 was tested by ELISA kit. Results: In the priming and early phase (0-48 h), or middle and later phase (48-96 h), or all phase (0-96 h) of mixing culture, the proliferation of T cells from experimental groups with addition of HVEM antibodies was higher compared with control groups (P<0.05), and the secretion of IL2 from CD3+T cell and CD4+ T cell groups was also higher compared with control groups (P<0.05). Conclusion: The negative signal produced by interaction between BTLA and HVEM played the inhibition role in T cell activation. Especially in the priming and early phase (0-48 h) of mixing culture of T cells and BMDC, the negative signal from BTLA already played such inhibition role, exhibiting another negative regulation mode different from traditional negative feedback regulation mode and suggesting the existence of multiple negative regulation for T cell activation.

294 Neurological effects of coexposure of dioctyl phthalate and arsenic in mice and associated mechanism

Objective: To investigate the neurological effect of exposure to individual and mixtures of dioctyl phthalate(DOP) and arsenic(As) in mice and to discuss resultant preliminary mechanisms associated with the effects. Methods: DOP (1 000 mg/kg,i.g.) and As (10 mg/L,p.o.) were administered individually and as mixtures to 4 groups of mice. After 8 weeks of exposure, the enzyme activity, learning and memory abilities and As content in brain of mice were assessed by commercial kits, Morris water mase test and VistaMPX Simultaneous ICPOES.The expression of Bax, Bcl2 and Caspase3 were performed by Western blot. Results: Mice demonstrated poorer learning and memory performance in exposure groups compared to control.The As level of brain in exposure groups were significantly increased (P<0.05). AChE and SOD activities decreased, however,MDA and NOS showed an increase in exposure groups compared to control. The expression of caspase3 and Bax were significantly increased but Bcl2 expression was inhibited in the hippocampus, and Bax/Bcl2 increased. Conclusion: Both DOP and As can induce neurotoxicity in mice associated with hippocampal apoptosis which caused by oxidative stress. The joint toxicity of DOP and As was complicated. Effect of antagonism, synergistic or without interaction were all observed. The mechanism needs to be further studied.

299 Expression of ADAM17 and its effect on proliferation and migration in pancreatic cancer cell

Objective: To investigate the expression of a disintegrin and metalloproteinase17(ADAM17) of human pancreatic cancer cell and it′s effect on proliferation and migration in human pancreatic cancer cell. Methods: The protein and mRNA expression of ADAM17 in three human pancreatic cancer cell were detected by using quantitative realtime PCR and Western blotting.The inducible lentiviral vectors mediated short hairpin RNA(shRNA) interference targeting ADAM17 gene were constructed,and transfected it into pancreatic cancer cell line,then screened stably transfected clonal cell line.After different doxycline concentration to induce the expression of ADAM17shRNA,cell proliferation and migration were analyzed by using the Cell Counting Kit8 (CCK8) viability assay and scratch assay. Results: ADAM17 protein and mRNA were highly expressed in PANC1,SW1990 and Patu8988 human pancreatic cancer cells.After small interfering RNA (siRNA) to block the expression of ADAM17,the protein levels of ADAM17,cell proliferation and migration in the experiment group were significantly lower than that of control group(all P<0.05). Conclusion: ADAM17 protein and mRNA have been expressed highly in human pancreatic cancer cell and promotes the proliferation and migration of human pancreatic cancer cell.

304 Influence of long noncoding RNA UCA1 on invasion and metastasis of pancreatic cancer cell line PANC-1

Objective: To investigate the expression of urothelial carcinoma antigen 1 (UCA1) in pancreatic cancer cell lines and its influence on the invasion and metastasis of the pancreatic cancer cell line PANC-1. Methods: The expression level of UCA1 in 4 pancreatic cancer cell lines was detected by real-time PCR. PANC-1 cells were randomly divided into Vector group and UCA1 group,which was transfected with Vector and UCA1-plasmid, respectively; the UCA1 expression level in two groups was compared. The ability of migration and invasion in vitro in Vector group and UCA1 group were measured by Transwell migration assay and Transwell invasion assay. The protein level of MMP14, MMP2 and MMP9 in two groups was analysed by Western blotting. Results: UCA1 differentially expressed in 4 pancreatic cancer cell lines. Compared with the Vector group, the ability of migration and invasion in UCA1 group were dramatically enhanced (P<0.01), and the protein level of MMP14, MMP2 and MMP9 in PANC-1 were increased (P<0.01). Conclusion: Up-regulation of UCA1 promotes the invasion and metastasis in vitro of PANC-1.

308 Comparison of method for GITRL SNP-rs2236876 genotyping

Objective: Comparing three single nucleotide polymorphism (SNP) genotyping methods, to choose an accurate and effective SNP genotyping method for rs2236876A/G of GITRL gene. Methods: SNP genotyping for rs2236876A/G of GITRL were performed and compared by Taqman-MGB probe, the PCRRLFP and direct sequencing. Results: PCR-RLFP showed the genotyping result by electrophoresis, which was suitable for a small number of experimental subjects. Taqman-MGB probe showed the genotyping result by fluorescence, which was suitable for a large number of experimental subjects. Direct sequencing method was operated by the sequencing company which was expensive. Conclusion: For rs2236876A/G of GITRL gene, Taqman-MGB probe method could be chosen for genotyping, PCR-RFLP method confirmed the genotyping results and, at the same time, the direct sequencing finally validated the result of PCR-RFLP.

311 Mechanism of miRNA-145 inhibits cell proliferation of A549 cells

Objective: To investigate the mechanism underlying how miR-145 inhibits cell proliferation in A549 cells. Methods: The putative miR-145 binding sites in the 3′-UTR region of Sp1 gene were predicated by bioinformatics and was predicted by a luciferase method. miR-145 levels were measured by qPCR and Sp1 protein levels were measured by Western blotting after transiently transfected with either miR-145 mimics or inhibitors into A549 cells. MTT assays were employed to evaluate cell proliferation in A549 cells simultaneously at 24,48 and 72 h after transfection. Results: Through report of luciferase analysis, the study successfully predicted and verified the putative miR-145 binding site in the 3′-UTR of human Sp1 gene. The luciferase activity of the wild type is reported that it could be significantly reduced and induced 48 h post-transfection of miR-145 mimics and inhibitors respectively and it had significant difference between the above two groups(P<0.05). Compared to the control, transfection of miR-145 mimics or miR-145-inhibitors reduced or induced Sp1 protein levels with the significant difference(P<0.05). Data from real-time qPCR indicated that transfection of miR-145-mimics and inhibitors induced and reduced miR-145 levels respectively compared to the negative control with a significant difference(P<0.05). There was no significant difference between the negative control transfection group and transfection control group(P>0.05). Finally, transfection of miR-145-mimics and miR-145-inhibitor could significantly reduce and induce cell proliferation of A549 cells.Seventy-two hours post transfection, there had significant difference between miR-145-mimics transfection group and the negative control transfection group (P<0.05). Conclusion: miR145 inhibited cell proliferation of A549 cells through reducing Sp1 gene expression.

317 Plasmatic concentrations and the relationship with cardiac structure and function of TGF-β1 in patients with type 2 diabetes mellitus

Objective: To investigate the levels of serum concentrations of TGF-β1 in patients with type 2 diabetes mellitus, and to assess the potential association of TGF-β1 with cardiac structure and function. Methods: Seventy patients with type 2 diabetes mellitus (diabetes group) , and 50 healthy participants (control group) were enrolled in this study. Circulation concentrations of TGF-β1 were measured by means of ELISA. Cardiac index of each structure was measured by means of 2-dimensinal and M-model transthoracic echocardiograms. We evaluated mitral valve mouth blood flow spectrum by pulse doppler,and tissue Doppler imaging was used to measure mitral valve ring motion spectrum. Differences between the two groups were compared; and analyzed the correlation index between TGF-β1 and the indicators. Results: Compared with the participants in the control group, the thickness of LAD, IVSTd, LVPWTd in patients with diabetes was significantly thicker(P<0.01); and the level of LVM, and LVMI was significantly higher(P<0.01). E peak and E′ peak in patients with diabetes was decreased(P<0.01), but the E/E′ ratio was higher than that in healthy participants(P<0.01). Plasmatic concentrations of TGF-β1 and BNP levels were higher in the diabetes group compared with those in the control group(P<0.01). Multiple regression analyses revealed that LVM associated with TGF-β1 levels independently(β=0.219, 95%CI:0.012-0.158,P=0.023). Conclusion: Plasmatic TGF-β1 level increased significantly in the patients with type 2 diabetes compared with the healthy people; TGF-β1 levels in patients with type 2 diabetes might be available as predictors of ventricular remodeling, to provide a new basis for early diagnosis of diabetic cardiomyopathy.

322 Analyse the dosimetric difference and evaluate short-term clinical efficacy between VMRT and conventional 4-fields radiotherapy in cervical carcinoma

Objective: To analyse the dosimetry difference and evaluate recent clinical efficacy and acute toxicity reaction between volume of rotating intensitymodulated radiotherapy(VMRT) and conventional pelvic 4-fields radiotherapy(4F) in advanced cervical carcinoma(stage Ⅱb-Ⅳ). Methods: There were 32 patients of advanced cervical cancer patients with newly diagnosed,and we designed both VMRT and 4F radiotherapy plan after CT simulation for all of the patients, evaluated the dosimetry of target volume and the organs at risk. The patients were divided into VMRT group(n=15) and 4F radiotherapy group(n=17) by the willingness of patients,VMRT(50 Gy/28 f) or 4F(45-50 Gy/25-28 f) radiotherapy carried out combined with brachytherapy and concurrent cisplatin chemotherapy(40 mg/m2,QW),and then compare the short term effect and acute toxicity reaction. Results: Both of uniformity(HI) and conformal(CI) of target volume in the VMRT group were better than that in the 4F group(P<0.05).In the VMRT plan,V30,V40, Dmean,D50,V40 and Dmean of the small intestine, bladder and rectum, were lower than that in 4F radiotherapy(P<0.05). Complete remission rate were 86.7% (13 cases) and 82.4%(14 cases, P>0.05) in VMRT group and 4F radiotherapy group respectively. Radiation induced enteritis and cystitisand were lower than that in 4F radiotherapy group(P<0.05).Both groups showed varying degrees of bone marrow suppression,but there was no statistically significant difference(P>0.05). Conclusion: In dosimetric analysis,VMRT showed advantage in the protection of organs, there was no difference between two group in the short term effect, but the incidence of acute toxicity significantly reduced.Above all,VMRT will be recommended for the patients with cervical carcinoma who received external beam irradiation.

326 Correlation between arterial stiffness and left ventricular diastolic function in prehypertension

Objective: To study the correlation between left ventricular diastolic function and aortic stiffness in prehypertension. Methods: A total of 40 cases of untreated prehypertension as observation group and 40 pure normaltensive cases as control group.Left ventricular diastolic function was assessed by echocardiography, aortic distensibility parameters were derived from aortic diameters measured by ultrasonography, and vascular inflammation was assessed by high-sensitivity C-reactive protein. Results: Prehyperten-sive subjects displayed a significantly lower E/A ratio and a significantly greater deceleration time and isovo-lumetric relaxation time compared with normoltensive controls. They also displayed aortic systolic diameter, diastolic diameter and mean aortic stiffness significantly increased while systo-diastolic diameter change, mean aortic distensibility and aortic strain were significantly reduced compared with controls. Values of infammatory markers were increased. At multiple regression analysis, E/A ratio was significantly related to highsensitivity C-reactive protein(β=0.002,P=0.019) and aortic stiffness(β=-0.128,P=0.028) after correction for age, left ventricular mass index and mean blood pressure. Conclusion: In prehypertension,the failing of left ventricular diastolic function and aortic stiffness have been started, showed significant correlation.

331 Expression and significance of serum IL-23 in patients with Hashimoto′s thyroiditis

Objective: To determine the levels of plasma interleukin-23(IL-23) and interleukin-17(IL-17), and analyze the clinical significance of them in patients with Hashimoto′s thyroiditis(HT). Methods: Enzyme linked immunosorbent assay (ELISA) was used to analyze the concentration of plasma IL-23 and IL-17 in 61 HT patients and 37 healthy controls. Results: The concentration of plasma IL-23 and IL-17 was significantly elevated in the HT patients than the controls(P<0.01).A positive correlation was observed between the level of IL-23 and IL-17. The level of plasma IL-23 in hypothyroid patients with HT showed positive correlations with the level of serum TGAb or TPOAb, but the correlation between the level of plasma IL-23 in euthyroid patients and the level of serum TGAb or TPOAb were not found. Conclusion: IL-23 may play an important role in the pathogenesis of HT.

335 Glucose metabolism level changes in neural circuits of the first-episode single-phase major depressive disorder patients using PET-CT

Objective: To study the changes of glucose metabolism in neural circuits of the first-episode single-phase major depressive disorder patients changes of glucose metabolism. Methods: Using PET-CT to scan 30 first-episode single-phase major depressive disorder patients and 30 healthy persons, injecting 0.2 mCi of 18F-FDG per kilogram of body weight,detecting the SUVmax value of the neural circuits respectively (including hippocampus, thalamus, frontal lobe and cingulate gyrus). Results: Compared with the healthy group, the SUVmax of bilateral hippocampus ,thalamus ,frontal lobe and cingulate gyrus in the first-episode single-phase major depressive disorder patients were reduced significantly(P<0.001 or P<0.05); the thalamus changes more obviously. Conclusion: The nerve cells in neural circuits of the first-episode single-phase major depressive disorder patients have been damaged,it was reflected in the intake of energy utilization, and the thalamus displayed more obviously.

339 Change of carotid morphology and hemodynamics in patients with essential hypertension

Objective: To explore the relationship between essential hypertension (EH) and carotid athe-rosclerosis, observing carotid artery morphological and hemodynamic condition by color Doppler ultrasound in patients with EH. Methods: A total of 48 hypertensive patients were selected as the observation group and 62 healthy volunteers during the corresponding time in our hospital for medical examination were as the control group. Doppler ultrasound was used to measure every patient′s carotid artery diameter (D), inside-media thickness (IMT), diameter stenosis rate(D′), peak systolic velocity(PSV), end-diastolic velocity(EDV), peak systolic velocity/end-diastolic velocity (PSV/EDV), resistance index and pulsatility index; and calculate the maximum shear rate (SR) and plaque detection rate of each group. Finally these data were analyzed by SPSS 19.0 software package. Results: Obviously, the observation group′s D, IMT, D′, PSV/EDV, resistance index and pulsatility index were higher, while PSV, EDV and SR were lower than the control group. Besides, the plaque detection rate of the observation group was also markedly higher than the control group. All of these differences were statistically significant (P<0.05). Conclusion: EH is likely to affect the carotid vascular morphology and hemodynamics and is an independent risk factor in the carotid atherosclerosis. In the process of carotid atherosclerosis, hypertension plays an important role in the carotid artery by the synergistic effect of high and low shear.

342 Nested fluorescent PCR combined with high resolution melting curve to detect and distinguish Mycobacterium tuberculosis strains

\[Abstract\]Objective: To establish a PCR method for detection of Mycobacterium tuberculosis strains. Methods: According to the TtoC transition at position 215 prior to the start codon of nitrate reductase gene(narGHJI) between Mycobacterium tuberculosis and Mycobacterium bovis, a nested fluorescent PCR combined with a high-resolution melt assay was designed. The sensitivity of the method was determined by comparing the quantitative results with commercial quantitative reagent using Mycobacterium bovis BCG as specimen, and the PCR products were observed by agarose gel electrophoresis to verify the specificity. Then, the method was applied to analyze the normalized melt curves and melting temperatures of the PCR product among Mycobacterium tuberculosis (H37Ra), Mycobacterium bovis BCG, clinical isolates of Mycobacterium tuberculosis and bovis. Results: narGHJI in Mycobacterium tuberculosis was detected specifically by nested fluorescent PCR, and the sensitivity was higher than compared method about 10 fold. By analyzing the melting curves of the PCR product for Mycobacterium tuberculosis using high-resolution melt assay, Mycobacterium tuberculosis and Mycobacterium bovis was divided by each other into two different combinations, and the melting temperature for Mycobacterium tuberculosis (83.08±0.08)℃ was significantly lower than Mycobacterium bovis (83.96±0.09)℃(t=217.23,P<0.05). Conclusion: A nested fluorescent PCR combined with high-resolution melt assay was established successfully to detect and distinguish Mycobacterium tuberculosis strains.

346 Signaling lymphocytic activation molecule expression on CD4+ T cells surface in patients with allergic rhinitis

Objective: To explore the changes of signaling lymphocytic activation molecule (SLAM) expression on CD4+T cells surface in patients with allergic rhinitis and the relationship between SLAM expression and the disease state. Methods: Eighteen patients with allergic rhinitis and sixteen healthy controls were enrolled in the study. PBMC were isolated from the whole blood by Ficoll gradient centrifugation. The expression of SLAM on CD4+ T cell surface was assessed by using flow cytometry, the expression of plasma IgE was tested by ELISA. Then we analyzed the correlation between SLAM expression and plasma IgE level. Results: Compared with the healthy controls, the expression of SLAM on CD4+ T cell surface was significantly decreased in allergic rhinitis patients(t=2.29,P<0.05), but the plasma IgE level was significantly increased(t=2.740, P<0.01); and there was a negative correlation between SLAM expression and plasma IgE level(r=-0.484 8,P<0.05). Conclusion: Patients with allergic rhinitis showed low expression of SLAM in CD4+T cells in peripheral blood, which might reflect the disease severity to some extent.

349 Preparation of hearttype fatty acidbinding protein and cardiac troponinⅠ bivalent strip

Objective: To develop the bivalent strip of human hearttype fatty acid binding protein(HFABP) and cardiac troponinⅠ(cTnⅠ) using colloidal gold immunochromatography technology with principles of doubleantibody sandwich assay,for the early and midassisted diagnosis of acute myocardial infarction. Methods: The colloidal gold was prepared by trisodium citrate reduction method, and its related factors were investigated. Colloidal gold particles labelled with monoclonal antibody of HFABP and cTnⅠ,respectively,were coated on some glass fiber and dried; similarly, its factors were optimized. The other monoclonal antibody of cTnⅠ,HFABP and goat antimouse IgG were blotted on the test one line,test two line and control line of nitrocellulose membrane, which were prepared with the absorbent pad,the pretreated sample pad and PV floor into a bivalent strip. Results: The test strip of HFABP and cTn Ⅰ detection sensitivity was 10 ng/mL and 3.0 ng/mL, respectively, the result can be determined within 10 min, that have uniform speed chromatography, no background, no crossover between the two proteins response and good stability. Conclusion: It prepared HFABP/cTnⅠ colloidal gold bivalent strip of high sensitivity,uniform speed chromatography and good stability,successfully.

356 Guidance of thromboelastography and blood coagulation indexes in rational use of blood components to HBV patients

Objective: To discuss the guidance of thromboelastography (TEG) and blood coagulation indexes in rational use of blood components to HBV patients. Methods: To compare the parameters from 128 patients under the guidance of TEG detection and blood coagulation indexes, and to observe and analyze the actual bleeding conditions. Results: APTT, PT, INR and R, K had a positive correlation, the r values were 0.385, 0.440, 0.423 and 0.437, 0.300, 0.290, P<0.05. The abnormal rate of R (19.53%) was significantly reduced compared to INR (32.81%),χ2=3.21,P<0.05; the abnormal rate of MA (15.63%) was significantly reduced compared to PLT (25.78%) too, χ2=2.62,P<0.05. Conclusion: The jointly monitor of TEG detection and blood coagulation indexes in rational use of blood components for HBV patients may have guiding significance.
