Abstract:Objective: To observe the effects of MTA1 small interfering RNA(siRNA)on anchorage-independent growth and anoikis of bladder cancer cell line BIU-87. Methods: After construction of the MTA1 gene eukaryotic expression vector of siRNA and transfected to the bladder cancer cell line BIU-87, the expression of MTA1 gene was detected by real-time PCR and western blot; to detect anchorage-independent growth using clon formation in soft agar,and anoikis using DNA fragmentation assay and flow cytometry assay,respectively. Results: The level of mRNA and protein of MTA1 of groups transfectd with siRNA were decreased markedly in a time-and dose-dependent manner. MTA1 siRNA can inhibit anchorage-independent growth in a dose-dependent manner. MTA1 can reduced the resistance to anoikis of BIU-87 cells in a dose-and time-dependent manner. Conclusion: MTA1 siRNA can inhibit the growth of bladder cancer cell line BIU87 through inducing anoikis.
