Abstract Objective: To investigate the effect of methyl-CpG-binding protein 2 (MeCP2) on the proliferation and migration of pancreatic cancer cell line PANC1. Methods: We first detected the MeCP2 expression in PANC1, PaTu8988 and SW1990 pancreatic cancer cells using qRT-PCR and Western blotting. Then plasmid shEGFP, shMeCP2 were transiently transfected into PANC1 cells, named as control group and experiment group, respectively. The relative proliferation rates were examined by CCK-8 and colony formation assays. The relative migration rates were examined by Transwell assays. Results: The expression of MeCP2 was different in three pancreatic cancer cell lines. Compared with the control group, MeCP2 mRNA and protein level was decreased(P<0.05) and the ability of proliferation, cell colonyforming and migration were significantly inhibited in PANC1 cells in experiment group (all P<0.05). Conclusion: Knockdown of MeCP2 could decrease the proliferation and migration of PANC1 pancreatic cancer cells.
