Analysis of the major metabolites of Sijunzi decoction in rat serum based on HPLC-MS |

Abstract Objective: To determine the decoction and drug-containing serum of Sijunzi decoction by high performance liquid chromatographymass spectrometry(HPLC-MS), and analyse the major metabolites of Sijunzi decoction. Methods: The decoction was prepared according to the proportion of traditional Chinese medicinal materials of sijunzi decoction, and the blood was collected 2 h after intragastric administration Sijunzi decoction to 6 rats. HPLC-MS was used to identify the active components in decoction and drug serum. Results: According to the total ion chromatogram obtained by HPLC-MS and the fragmentation peak of mass spectrometry, it was deduced that Sijunzi decoction contained 10 chemical substances, among them, 7 saponins were ginsenoside Rb1, Re, Rg1, Rb2, Rd, Rg3, glycyrrhizic acid; 3 flavonoids were liquiritin, glycyrrhizin, licorice coumarin. Drug-containing serum contains three prototype chemical composition, which are ginsenoside Rb1, Re, glycyrrhizic acid and 2 transitional components, glycyrrhetinic acid and glycyrrhizin glucuronide. Conclusion: The chief chemical compositions of the decoction and drug-containing serum of Sijunzi decoction were ginsenosides and glycyrrhiza, the former was metabolized in blood by prototype compound while the latter was metabolized in blood by hydrolysis product.
