Determination of monosaccharides composition of Cistanche tubulosa polysaccharides by gas chromatography |

Abstract Objective: To explore the optimal methods for analysis of the monosaccharides composition of Cistanche tubulosa polysaccharide. Methods: To investigate modifying factors of the polysaccharide hydrolysis was determined by single factor method, and the monosaccharide composition analysis of Cistanche tubulosa polysaccharide by gas chromatography. Results: The optimal hydrolysis conditions were sulfuric acid concentration 3.0 mol/L, hydrolysis time 8 h and temperature 110 ℃;the RSD value of precision was between 0.92% and 4.68%,the RSD value of repeatability was between 0.92% and 1.77%, the RSD value of stability was between 1.61% and 3.61%, the recovery rate were between 99.3% and 100.3%; Cistanche tubulosa polysaccharide was mainly composed of mannose, glucose and galactose in a molar ratio of 2.01 ∶5.72 ∶2. Conclusion: The refined method can be used for monosaccharides composition analysis of Cistanche tubulosa polysaccharides.
