Effect of PECAM-1 siRNA on cell proliferation and apoptosis of irradiation-induced drug resistance CNE1/R cells of human

Abstract [Abstract]Objective: To investigate the effect of silencing platelet-endothelial cell adhesion molecule-1(PECAM-1) by small interfering RNA (siRNA) on cell proliferation and apoptosis of irradiation-induced drug resistance CNE1/R cells of human nasopharyngeal carcinoma. Methods: The CNE1/R-specified siRNA was designed and synthesized. The purity of the transfected cells were screened out by G418. The expression of PECAM-1 was detected by western blotting. Cell growth and apotosis was assayed by MTT and flow cytometry, respectively. Results: The expression of PECAM-1 protein in CNE1/R cells was significantly downregulated. Compared with negative group and control group, experiment group showed significantly decreased proliferation ability in CNE1/R cells, whereas cell apoptosis rate was increased. Conclusion: PECAM-1 siRNA could inhitit the growth and promote the apoptosis of CNE1/R cell, which suggested PECAM-1 might play a key role in CNE1/R cell growth and apoptosis.
