Preventive effect of bFGF intradiscal injection on rats′ lumbar disc degeneration |

Abstract Objective: To observe the effect of prevention of SD rat lumbar disc acupuncture degeneration by the intradiscal injection of basic fibroblast growth factor (basic fibroblast growth factor, bFGF).Methods: Twenty-four SD rats were randomly divided into test group,control group and blank control group. There were 8 rats in each group. The posterior annulus fibrosus of L4/5 andL5/6 disc of test group were stabbed by 27-gauge hypodermic needle, then used the micro-injection syringe to inject 50 μL diluted bFGF. 50 μL saline was injected into the discs of control group. The discs of blank control group weren't stabbed. Eight weeks after surgery,all the rats were examined by MRI, the T2 weighted signal intensity of each group was measured under MRI. The states of disc degeneration were scored by Pfirrmann grading. Then all the rats were sacrificed and the discs were removed for pathological observation. Results: MRI showed signal intensity of nucleus pulposus in test group and blank control group was significantly higher than that in control group 8 weeks after surgery which resulted in a significant Pfirrmann score difference(P<0.05) among groups. Pathological examination showed the number of nucleus pulposus cells of test group and blank control group had no significant change and the fibers arranged in neat rows, meanwhile the number of control group nucleus pulposus cells significantly reduced, and the disordered arrangement of collagen fibers was also observed. Conclusion:Intradiscal injection of bFGF had a preventive effect for the SD rat intervertebral disc degeneration after posterior annulus fibrosus acupuncture.
