Abstract Objective: To study the mitochondria mechanism of olfactory ensheathing cells conditioned medium(OECCM) against H2O2 induced injury in astrocytes. Methods: After treatment of 500 μmol/L H2O2 for 20 min, 50% OECCM was added to treat the cells for 24 h; MTT assay was used to analyse astrocytes viability; apoptosis induced by 500 μmol/L H2O2 was decreased by hoechst 33342 staining and the expression of caspase-3 by Western blot; mitochondrial membrane potential was detected by JC-1 staining. Results: OECCM could protect astrocytes from the injury of 500 μmol/L H2O2; apoptotic cells were decreased and the expression of caspase-3 was decreased also; OECCM could increase mitochondrial membrane potential. Conclusion: OECCM could against H2O2 induced injury in astrocytes. Its mechanism might be related to stabilize mitochondria function.
