EGCG对甲状腺癌细胞增殖和摄碘能力的影响

摘要目的: 观察表没食子儿茶素没食子酸酯（epigallocatechin3gallate, EGCG）对甲状腺癌细胞增殖和摄碘能力的影响。方法: 用不同浓度的EGCG处理甲状腺癌W3细胞株48 h后，用3(4，5二甲基噻唑2)2，5二苯基四氮唑溴盐（MTT）法检测W3细胞增殖能力，流式细胞术、免疫荧光法检测钠碘转运体（sodium iodide symporter，NIS）的变化，摄碘能力实验检测W3细胞摄碘能力。结果：与对照组比较，随着EGCG浓度的增加，W3细胞的增殖能力逐渐降低（P<0.01），NIS的表达逐渐增加（P<0.01），呈浓度依赖性。摄碘实验显示，在一定浓度范围内，EGCG明显促进W3细胞对碘的吸收能力 (P<0.01)。结论： EGCG能抑制甲状腺癌细胞的增殖能力，通过上调NIS的表达促进甲状腺癌细胞株的摄碘能力。

Abstract：Objective: To explore the effect of epigallocatechin-3-gallate(EGCG) on the proliferation and radioiodine accumulation in papillary thyroid cancer(PTC) cells. Methods: After PTC cells W3 were treated by using different concentrations of EGCG for 48 h, the proliferation of PTC cells was detected by 3-4,5dimethylthiazol-2-yl-2,5-diphenyl-tetrazolium bromide（MTT） assay; the expression of sodium iodide symporter(NIS) in PTC cells was detected by flowcytometry and immunofluorescence methods; the ability of PTC cells to iodide-uptake was measured by iodide-uptake assay. Results: The proliferation of PTC cells was decreased, and the expression of NIS in PTC cells was increased, in a concentration dependent manner.And the ability of iodideuptake in PTC cells was increased significantly in some concentration gradient. Conclusion: EGCG inhibited the growth of PTC cells and enhanced the ability of iodideuptake through increased expression of NIS.
