Abstract The optimization chromatography analyses of puerarin and daidzin content were investigated under various conditions of the selections for TLC solvent and coloration, HPLC chromatographic separation conditions and detection characteristics, and MEKC chromatographic separation conditions and detection characteristics. The results show that the correctness and precision of MEKC are slightly lower than those of HPLC with lower sensitivity of daidzin, while it is worth further studying due to the low organic solvent consumption, quick peak flowing out and environmental friendly characteristics. Isoflavones of puerarin and daidzin can be rapidly identified by TLC with better separation efficiency than that in developing solvent of pharmacopoeia method. Compared with previously report, HPLC has lower detection limit and wider detection range, and at least 5 alcohol extractive components can be separated well from impurities with quantitative analyses of puerarin and daidzin. The rapidly and accurately qualitative and quantitative detecting of puerarin and daidzin in radix pueraria can be realized by TLCHPLC under the optimal conditions.
