Effect of rapamycin inhibit liver fibrosis in rat with obstructive jaundice |

Abstract Objective: To study the effect of rapamycin on liver fibrosis in rats with obstructive jaundice, and to explore its mechanism. Methods: The common bile duct was ligated in rats with obstructive jaundice model, 36 SD rats were randomly divided into rapamycin treatment groups (treatment group)(n=18), and the only obstructive jaundice(control group) (n=18). The endotoxin content of liver was monitored each time, while 6 rats of each group were sacrificed on postoperative day (POD)1, 3 and 5,and hepatic tissue samples were harvested to determine TNF-α protein expression levels, with observing the pathological changes of the liver tissue with light microscope.Results: Compared with the control group, the endotoxin level and TNF-α protein expression decreased significantly in treatment group(P<0.01). The main pathomorphology changes were intrahepatic bile duct proliferation and hepatic fibrosis. In the same time point, pathological changes of the liver tissue was significantly different between the control group and the treatment group. Conclusion: The rapamycin can inhibit liver fibrosis with obstructive jaundice in rats through inhibiting the JAK/STAT signaling pathway.
