Abstract:Objective: To establish a HPLC method for simultaneous determination of caffeic acid, pcoumaric acid, scopoletin, rosmarinic acid, esculetin, rutin, hyperin and quercetin in Prunella vulgaris L.. Methods: The separation was performed on a InertSustainTM ODSC18 column (250.0 mm×4.6 mm, 5 μm), with mobile phase consisted of acetonitrile-0.1% acetic acid solution flowing at 1.0 mL/min in a gradient elution manner. The column temperature was 30 ℃ and the detection wavelength was set at 325 nm and 360 nm. The established HPLC method was investigated by linear relationship test, precision test, stability test, repeatability test and recovery test. Results: The eight components showed good linear relationship in the range of 1-100 μg/mL, and the average recoveries were from 98.63% to 102.36%. Relative standard deviations of precision test, stability test and repeatability test were all lower than 3%. Conclusion: This simple and reliable method may be applied to accurately determine the contents of eight components, which could be used for the quality control of Prunella vulgaris L..
