Expression of microRNA-187 in gastric carcinoma and its effect on proliferation and invasion of gastric cancer cells

Abstract:jective: To investigate the effect of abnormal expression of microRNA187(miR187) on proliferation, invasion and migratory of gastric cancer. Methods: A total of 24 pairs of human gastric cancer tumor and matched nontumor tissues were collected and miR187 expression was detected and their relevance was analyzed. The expression of miR187 and CD276 in human gastric cancer cell SGC7901 were inhibited by siRNA transfection, and then the effect of SGC7901 cell proliferation and invasionmigration was detected by CCK8 and Transwell assay respectively. The expression of matrix metalloproteinase9(MMP9) was detected by Western blotting. Results: Compared with matched nontumor tissues, the expression of miR187 was increased in stage Ⅰ,Ⅱ,Ⅲ and Ⅳ gastric cancer(P<0.01); the expression of CD276 was decreased in stage Ⅰ,Ⅱ,Ⅲ and Ⅳgastric cancer(P<0.01). The correlation coefficient of miR187 expression and CD276 expression in gastric carcinoma was 0.840. Compared with group miR187 inhibitorc, the expression of CD276 was upregulated and MMP9 was downregulated(P<0.01), the proliferation of SGC7901 and the invasion and migration of SGC7901 was inhibited(P<0.01) in group miR187 inhibitor. Compared with group siRNAnegative control, the expression of MMP9 was upregulated in the group CD276siRNA (P<0.01). Conclusion: MicroRNA187 was increased in gastric carcinoma and the proliferation and invasion of human gastric cancer cell was promoted by miR187.
