Abstract:Objective: To investigate the effects of YAP knockdown in gastric cancerderived mesenchymal stem cell(GCMSC) on proliferation, migration and angiogenesis of gastric cancer. Methods: For YAP knockdown, the lentiviralYAP were infected into GCMSC. qRTPCR and Western blotting were used to test the efficacy of YAP knockdown. The mRNA expression of VEGF, PDGF and IL8 were tested by qRTPCR. By collecting the conditioned medium(CM) of GCMSC YAP shRNA group and control group, the change of angiogenesis was detected by using the endothelial tube formation assay. Colony formation assay and cell migration assay were used to test the proliferation and migratory ability of SGC7901 treated with the CM of YAP knockdown GCMSC. Results: Compared with the control group, the protein and gene expression levels of YAP decreased significantly (P<0.01). The decreased expression of vascular endothelial growth factor (VEGF), platelet derived growth factor(PDGF) and IL8 were also observed (P<0.01). The angiogenesis ability of YAP knockdown GCMSC decreased significantly. YAP knockdown in GCMSC could inhibit the proliferation and migration of SGC7901. Conclusion: YAP knockdown in GCMSC inhibited angiogenesis and reversed the promotion of GCMSC on the proliferation and migratory of SGC7901. It could be implicated that YAP may be involved in the angiogenesis, proliferation and migration of gastric cancer cells.
