Expression and significance of TGFβ1, Smad3,Snail1 and FoxM1 in primary pterygium |

Abstract Objective: To observe the expressions of TGF-β1, Smad3, Snail1 and FoxM1 in the tissues of primary pterygium, and then to analyze the differences in the pterygium group compared with the pterygium-adjacent conjunctiva group. Methods: The expressions and ditributions of TGF-β1, Smad3, Snail1 and FoxM1 were examed by immunohistochemical staining of tissue sections of 30 pterygial tissues and 5 pterygium-adjacent conjunctival tissues. The positive staining rates of the epithelial cells for TGF-β1, Smad3, Snail1 and FoxM1 were calculated, and analyzed to show the differences in the pterygium group compared with the pterygium-adjacent conjunctiva group. Results: The hematoxylin-eosin staining of the tissue sections showed that the epithelia covering the pterygium-adjacent conjunctival tissues were composed of less layers of epithelial cells, which arranged regularly in the pterygium-adjacent conjunctiva group.There were fewer blood vesseles in the the pterygium-adjacent conjunctival lamina propria. The epithelia of the pterygium tissues were composed of more than ten layers of epithelial cells which distributed disorderly. Compared to the pterygium-adjacent conjunctiva, there were more blood vesseles in the lamina propria of the pterygium. The results of immunohistochemical staining of the tissue sections showed that there were fewer positive cells for TGF-β1, Smad3, Snail1 and FoxM1 in the epithelia of pterygium-adjacent conjunctival tissues; in the pterygium group, these positive cells were distributed extensively in the epithelia and substantia propria layer of the tissues. TGF-β1 expressed in the cytoplasm of the epithelial cells and the stroma cells in the substantia propria layer. The other proteins expressed in the nuclei of these cells distributing extensively in the pterygium tissues. With regard to the positive cells count in the pterygium-adjacent conjunctiva group and pterygium group, there were statistical significances(P<0.01) for the positive rates of TGF-β1, Smad3, Snail1 and FoxM1. Conclusion: The over-expressed TGF-β1, Smad3, Snail1 and FoxM1 in pterygium may play an important role in the genesis and development of pterygium, which may be induced by activiting the pathway of TGF-β1-Smad3-Snail1-FoxM1.
