Effect of CyPA on ox-LDL-induced activation and apoptosis in RAW264.7 cells |

Abstract [Abstract]Objective: To investigate the effect of cyclophilin A(CyPA) on ox-LDL-induced activation and apoptosis in RAW264.7 cells. Methods: RAW264.7 cells were incubated with 40 mg/L ox-LDL for 24 h, Western blotting analysis was employed to detect the CyPA expression. After transfected with CyPA siRNA, the cells were incubated with 40 mg/L ox-LDL for 24 h. The positive rate of CD80, CD86 and the apoptosis rate were determinated by flow cytometry analysis. Results: After incubating with ox-LDL, the level of CyPA was upregulated. The percentage of CD80 positive cells,CD86 positive cells and apoptotic cells were significantly decreased in CyPA knockdown cells compared with the negative control siRNA cells after the intervention of oxLDL. Conclusion: CyPA may mediate the oxLDLinduced activation and apoptosis in RAW264.7 cells.
