The expression characteristic of B7 costimulatory molecules
in human hematologic malignancy cell lines

Abstract:Objective: To investigate the expression and distribution characteristic of B7 costimulatory molecules in human hematologic malignancy cell lines. Methods: The expression and molecules subcellular distribution of B7.1, B7.2, HLA classⅠ, HLA classⅡ, and B7H1, CD279, B7H3 and B7H4 in 13 human hematologic malignancy cell lines were determined by flow cytometry, with the peripheral blood mononuclear cells(PBMCs) of 12 volunteers as the control. Results: Multiple human hematologic malignancy cell lines expressed low or undetectable level of B7.1 on the cell membrane, detectable or high level of B7.2, and high or detectable level of HLA classⅠand Ⅱ molecules. The membrane proteins of B7H1, CD279, B7H3 and B7H4 had low or no expression in cell lines, while the expression abundance of B7H3 was higher in U937, Maver and Z138 cell lines. The cytoplasmic protein of B7H1, B7H3 and B7H4 almost had no expression in cell lines, while the relative nuclear and cytoplasmic protein were abnormal expression or high expression in these cell lines, except for CZ1. Conclusion: There were significant differences among B7 molecules expression in both hematologic malignancy cell lines with different types, and the cell lines derived from the same type of tumor, which might be involved in the immune escape, the occurrence and development of hematological malignancies.
