Abstract:Objective: To investigate expression profile of dermatopontin(DPT) in various tissues, and different expression in adipose tissue of dietinduced obese mice and the effect of rosiglitazone on DPT expression in 3T3L1 adipocyte was also evaluated. Methods: qRTPCR and Western blotting were used to measure DPT mRNA and protein expression in major tissues of 6 weeksold mice, and also in adipose tissue of obese mice and control;the expression of DPT in stromal vascular fraction cells and adipocytes from adipose tissue was detected; 3T3L1 cell was induced to differentiate into adipocyte, which was treated with rosiglitazone at 0.25,0.5,1,2 μmol/L for 24 h, or at 2 μmol/L for 4,12,24,48 h, then DPT expression was measured. Results: DPT was highly expressed in adipose tissue, and obese mice showed higher expression of DPT mRNA and protein in adipose tissue compared with those of the control; DPT mainly expressed in the adipocyte; the expression level of DPT was gradually increased during the differentiation process of 3T3L1 cells; after treating with rosiglitazone for 24 h, the expression of DPT in adipocyte was decreased. Also DPT expression was also decreased after treating with 2 μmol/L rosiglitazone for 24 h and 48 h. Conclusion: Obesity obviously induces DPT expression in adipose tissue; rosiglitazone may regulate expression of DPT in adipocyte. It suggests that DPT may play a potential role in the development of obesity.
