Effect of treprostinil on expression of inhibitor of differentiation in rat model with pulmonary arterial hypertension

Abstract:Objective: To study the therapeutical effect of treprostinil in monocrotaline induced pulmonary arterial hytertention rats,and on the expressional levels of differentiation inhibitor 1 and 3(ID1,ID3). Methods: A total of 30 rats were randomly divided into:normal control,monocrotaline model,and treprostinil intervention groups(n=10 each). The pulmonary arterial hypertension model was developed with monocrotaline injection(60 mg/kg). The intervention group were injected with treprostinil for 2 weeks by implanted micro pump in the back, and the model group and the control group were given equal volume of normal saline.The mean right ventricular pressure(mRVP) were detected; the right ventricular hypertrophy index(RVHI) were measured by weighing each part of the heart;making lung tissue sections and HE staining,the percentages of wall thickness of pulmonary arteriole to external diameter of artery(WT) and vascular wall area to total vascular area(WA) were determined.Immunohistochemical method was used to detect the light density value of ID1 and ID3 in pulmonary vessels. Western blotting analyses and real-time PCR were employed to detect the ID1,ID3 and ID1 mRNA,ID3 mRNA expressions in lung tissue. Results: The mRVP, RVHI,WT and WA in both model group and the intervention group were significantly higher than those in control group(P<0.05),which suggested that the model of pulmonary hypertension was successfully established.The intervention group were lower than the model group(P<0.05) in mRVP,RVHI,WT and WA.Compared with the control groups, model group were lower in light density value of ID1, ID3 of pulmonary vascular,the ID1,ID3 and the ID1 mRNA and ID3 mRNA of lung tissue(P<0.05), and the above parameters of intervention group higher than those of the model group(P<0.05). Conclusion: Treprostinil decreases rat pulmonary arterial hypertension, and promotes the expression of ID1,ID3,ID1 mRNA and ID3 mRNA in lung tissue of rats with pulmonary arterial hypertension induced by monocrotaline.
