Abstract:Objective: To observe the effect of CO-releasing molecules-2(CORM-2) on the random narrow pedicle skin flap in the SD rats with ischemia-reperfusion injury(IRI),and discuss the possible mechanism. Methods: Established IRI models of random narrow pedicle skin flap in rats.A total of 36 SD rats were divided into two groups at random:CORM-2 group,models of random narrow pedicle skin flap and treated by CORM-2;NS group,models of random narrow pedicle skin flap and treated by normal saline(NS).Test the activity of NF-κ B by Western blotting. The content of malonaldehyde(MDA) in tissue of skin flap was detected by phenobarbitone acid;the activity of superoxide dismutase(SOD) in tissue of skin flap was detected by xanthine oxidase;the level of IL-6 and tumor necrosis factor-α(TNF-α) in blood serum was detected by ELISA;pathological changes were observed with light microscope in the 3rd, 5th, 7th day after operation;survival rate of skin flap was the average survival length of skin flap deciding practical length of skin flap. Results: ① The skin flap uncovered by the 3rd, 5th, 7th day,compared with NS group,the content of MDA in the skin flap of CORM-2 group lower(P<0.01),the activity of SOD significantly increased (P<0.01). ② The skin flap uncovered by 3rd, 5th, 7th day,compared with NS group,the level of IL-6 and TNF-α in blood serum lower(P<0.01). ③ Histopathology showed that abundant capillary were noticed by application of CORM-2. ④ CORM-2 can effectively restrain the NF-κ B activation in random narrow pedicle skin flap model. ⑤ The skin flap was uncovered by the 7th day,compared with NS group,survival rate of skin flap of CORM2 group significantly higher(P<0.01). Conclusion: CORM-2 effectively reduced the narrow pedicle skin flap of rats with IRI, and improved the survival rate of flap.
