Abstract In order to reveal the effect of water deficit on physiological characteristics of maize, variations of its photosynthesis and fluorescence, physical damage and protection mechanism at the seedling stage under water deficit were analyzed. The results show that stomatal closure induced by leaf ABA accumulation could decrease photosynthesis and fluorescence, but water use efficiency was increased under mild and moderate water deficit. The maximum photochemical efficiency of PSII was not affected under mild water deficit, which suggested that oxidative damage was avoided by dissipation of excess excitation energy. However, under moderate and severe water deficit, the protective mechanism was not sufficient for avoiding photo-damage. Thus, the damage to PSII occured. In addition, water deficit could also induce reactive oxygen species accumulation. Under mild water deficit, the activity of enzymatic antioxidants was increased and non-enzymatic antioxidative compounds were accumulated for alleviating physiological damages. However, the antioxidant activities of enzyme and non-enzymatic system were exceeded by reactive oxygen species accumulation. Thus, the level of membrane lipid peroxidation was increased. It is provided theoretical guidance for optimizing water use in maize according to its physiological characteristics.
